摘  要:本实验旨在研究催乳素(PRL)基因在小尾寒羊母羊不同组织中的表达规律。选择42月龄、体重(73.2±2.0)kg、体况良好的小尾寒羊空怀母羊5只,屠宰后采集垂体、下丘脑、肾、子宫、卵巢、淋巴和乳腺组织,用实时荧光定量PCR方法检测各组织中PRL mRNA的相对表达量。结果表明:PRL mRNA在小尾寒羊母羊不同组织中的表达量由高到低依次为:垂体、下丘脑、淋巴、肾、乳腺、卵巢、子宫,其中,PRL mRNA在垂体和下丘脑中的表达量极显著高于淋巴、肾、乳腺、卵巢和子宫,垂体中PRL mRNA的表达量极显著高于下丘脑,而淋巴、肾、乳腺、卵巢、子宫中PRL mRNA表达量差异不显著。This study was designed to investigate the expression of prolactin(PRL)gene in different tissues of the Smalltail Han ewes.Five nonpregnant Small-tail Han ewes aged 42 months,weighing 73.2±2.0 kg and in good body condition were selected.The pituitary,hypothalamus,kidney,uterus,ovary,lymph and mammary gland tissues were collected after slaughter and the relative expression of PRL mRNA in each tissue was detected by quantitative real-time PCR.The results showed that the expression levels of PRL mRNA in different tissues of Small-tail Han ewes were from high to low:pituitary,hypothalamus,lymph,kidney,mammary gland,ovary,uterus.The expression of PRL mRNA in pituitary and hypothalamus was significantly higher than that in lymph,kidney,mammary gland,ovary and uterus(P<0.01).The expression of PRL mRNA in the pituitary was significantly higher than that in the hypothalamus(P<0.01).There was no significant difference in the expression of PRL mRNA in lymph,kidney,mammary gland,ovary and uterus(P>0.05).
