摘  要:试验旨在研究热应激对通城猪血清生化指标及空肠组织热休克蛋白HSP70、HSP90 mRNA表达的影响。选取遗传背景相似、100日龄、体重(45±5)kg的通城猪30头,随机分为6组,每组5头,分别为对照组和热应激2、4、6、8、10 d(每天09:00—16:00)处理组。(28±1)℃下适应性饲养7 d,于第8天开始,对照组仍置于(28±1)℃条件下,处理组迅速升温至(38±1)℃,各组每天测定直肠温度、体表温度和呼吸频率,到相应天数屠宰采集样品,进行血清生化指标测定和空肠组织热应激蛋白HSP70、HSP90 mRNA表达水平检测。结果表明:热应激期间,每天14:00通城猪直肠温度和呼吸频率变化最明显,但表皮温度无显著变化;与对照组相比,热应激使血清总蛋白、白蛋白和球蛋白浓度显著降低,第4天达最低值,血清谷草转氨酶浓度在第4天和第10天显著降低,在第8天显著升高,血清碱性磷酸酶、乳酸脱氢酶、肌酸激酶浓度随热应激处理时间显著升高,第4天达峰值;空肠组织HSP70、HSP90 mRNA表达水平呈先升高后降低的趋势并分别在第4天达到峰值(P<0.05)。结果提示,热应激会使通城猪体温升高,呼吸频率增加并通过血清因子的代谢和HSP70、HSP90的表达来响应热应激的影响,在热应激第4天响应最强烈,第6天以后趋于稳定,说明通城猪在长期热环境下具有较好的调节能力。In order to investigate the effects of heat stress on serum biochemical indexes and HSP70,HSP90 mRNA expression in jejunal of Tongcheng pig.Thirty 100-day-old female Tongcheng pigs with similar genetic backgrounds and average initial body weight of(45±5)kg were randomly divided into six groups of 5.All pigs were firstly raised under(28±1)℃with ad libitum feeding for 7 days.Then control group was still raised under(28±1)℃,and the temperature of test groups was suddenly increased from(28±1)℃to(38±1)℃,for 2,4,6,8,10d(from 10:00 to 16:00 every day),respectively.After that,rectal temperature,epidermal temperature and respiratory rate of all pigs were measured daily,all pigs were slaughtered and samples were collected.The content of total protein(TP),albumin(ALB),globulin(GLO),aspartate aminotransferase(AST),alkaline phosphatase(ALP),lactate dehydrogenase(LDH),creatine kinase(CK)in blood serum and HSP70,HSP90 mRNA expression levels in jejunal were measured.The results indicated that the rectal temperature and respiratory rate of Tongcheng pigs changed most obviously at 14:00,but the epidermal temperature did not change significantly during the heat stress period.The contents of serum TP,ALB and GLO in Tongcheng pig blood serum was significantly decreased by heat stress(P<0.05)and reached the lowest value on the 4th day.Serum AST levels decreased significantly on the 4th and 10th day and increased significantly on the 8th day(P<0.05).The serum ALP,LDH and CK activity increased with the time passing of heat stress,and peaked at d 4,which was significantly higher than that of other groups(P<0.05).The HSP70,HSP90 mRNA expression of jejunum was increased at first and decreased subsequently,and peaked at d 4,respectively.The conclusion show that heat stress could increase the body temperature and respiratory rate of Tongcheng pigs,and response to heat stress through metabolism of serum factors and expression of HSP70 and HSP90.The response was strongest on the 4th day of heat stress,and tended to be stable after
