摘 要:目的探讨实时荧光定量聚合酶链反应(PCR)技术及细胞培养法在流感病毒检测中的应用价值。方法选取2013年1月至2018年12月阜新市哨点医院采集的4026份疑似流感咽拭样本作为研究对象,先后采用不用的检验方式进行检测,研究组采用实时荧光定量PCR法,对照组采用细胞培养法,比较两种检测方法对流感病毒携带者样本临床分型阳性率的检测效果。结果研究组检出阳性样本512份。其中,检出新甲H1型149份,对照组分离毒株62株;研究组H3亚型206份,对照组分离毒株109株;研究组B型157份,对照组分离毒株97株,各组均未检出H5、H7、H9亚型。研究组检出阳性样本总数与对照组毒株总数呈正相关,且新甲H1型、H3型、B型与毒株分别呈正相关(P <0.05)。研究组检出流感病毒阳性率为25.68%,对照组检出流感病毒阳性率为6.66%,差异有统计学意义(P <0.05)。结论实时荧光定量PCR技术对流感病毒检测的速度更快,灵敏度更高,具有较高的临床应用价值。Objective To investigate the value of real-time fl uorescence quantitative PCR and cell culture in the detection of infl uenza virus.Methods From January 2013 to December 2018,a total of 4026 suspected infl uenza cases were collected from sentinel hospitals in Fuxin city,used diff erent inspection methods for testing successively.The study group were given real-time fl uorescence quantitative PCR method,and control group were given cell culture method.The positive rates of clinical typing of infl uenza virus carriers were statistically and compared between the two groups.Results A total of 512 positive samples were detected,of which 149 were detected,62 strains were isolated from the control group,206 in the study group,109 strains isolated from the control group,157 in the study group and 97 from the control group,and H5,H7,H9 subtype was not detected in each group.The total number of the study group was positively correlated with the total number of the control group strains,and the new type H1,the H3 type,the B type and the strain were positive correlation(P<0.05).The study group detected the infl uenza virus positive was 25.68%,the positive rate of infl uenza virus in the control group was 6.66%,which was signifi cantly higher in the study group than that in the control group(P<0.05).Conclusion Real-time fl uorescence quantitative PCR can detect infl uenza virus with faster speed and higher sensitivity,and has high clinical application value.
