摘　　要：目的探究基质金属蛋白酶-8(MMP-8)基因多态性与主动脉夹层的关系。方法分析华中科技大学同济医学院附属同济医院收治的35例主动脉夹层患者和38例健康志愿者作为研究对象,分别作为疾病组和对照组。收集各研究对象外周静脉血5 mL,提取DNA并检测MMP-8基因rs35231465、rs61754773和rs77504671位点多态性,分析疾病组和对照组间等位基因分布、基因型分布的差异,分析单倍型分布差异,同时检测不同基因型的人群MMP-8基因表达差异。结果与对照组比较,疾病组MMP-8基因rs35231465位点等位基因G(77.1%vs.60.5%)和rs77504671位点等位基因C频率(52.9%vs.35.5%)更高,差异有统计学意义(P<0.05)。与对照组比较,疾病组MMP-8基因rs35231465位点GG基因型(62.9%vs.34.2%)和rs61754773位点TT基因型频率(54.3%vs.15.8%)较高,差异有统计学意义(P<0.05)。与对照组比较,疾病组MMP-8基因rs35231465、rs61754773和rs77504671位点ACT单倍型频率较低(6.3%vs.21.7%),GTC单倍型频率较高(31.3%vs.13.8%),差异有统计学意义(P<0.05)。与对照组比较,疾病组rs61754773位点显性模型CC+CT频率(45.7%vs.84.2%)、rs77504671位点显性模型TT+TC频率(60.0%vs.81.6%)和rs35231465位点隐性模型GA+AA频率均较低(37.1%vs.65.8%),差异有统计学意义(P<0.05)。MMP-8基因rs35231465位点多态性与MMP-8基因表达明显相关,疾病组AA基因型患者MMP-8基因表达明显较高。结论MMP-8基因rs35231465位点GG基因型和rs61754773位点TT基因型与主动脉夹层发病易感性有显著关系。Objective To explore the relationship between matrix metalloproteinase-8(MMP-8)gene polymorphism and aortic dissection.Methods Thirty-five aortic dissection patients and 38 healthy volunteers in our hospital were taken as the research objects,respectively as disease group and control group.Five mL of peripheral venous blood of each study subject was collected,DNA was extracted and the polymorphisms of MMP-8 gene rs35231465,rs61754773 and rs77504671 were detected,and the differences in allele distribution and genotype distribution between the disease group and the control group were analyzed.At the same time,differences in MMP-8 gene expression in people with different genotypes were detected.Results The distribution of alleles of MMP-8 gene the allele G in rs35231465(77.1%vs.60.5%)and the allele C in rs77504671(52.9%vs.35.5%)in the disease group were more frequent high than those in the control group and the differencec were statistically significant(P<0.05).The frequency of the GG genotype at the rs35231465 locus(62.9%vs.34.2%)and the TT genotype at the rs61754773 locus(54.3%vs.15.8%)of the MMP-8 gene in the disease group were higher than those in the control group,and the differencec were statistically significant(P<0.05).The frequency of ACT haplotypes at rs35231465,rs61754773 and rs77504671 of MMP-8 genes in the disease group was lower(6.3%vs.21.7%),and the frequency of GTC haplotypes was higher(31.3%vs.13.8%)than those in the control group.The differences were statistically significant(P<0.05).The frequency of rs61754773 locus dominant model CC+CT(45.7%vs.84.2%),the frequency of rs77504671 locus dominant model TT+TC(60.0%vs.81.6%)and rs35231465 locus recessive model GA+AA(37.1%vs.65.8%)in the disease group were lower than the control group,and the difference was statistically significant(P<0.05),and the differences were statistically significant(P<0.05).MMP-8 gene rs35231465 locus polymorphism was significantly related to MMP-8 gene expression,and MMP-8 gene expression was significantly higher in patients wi
