丙烯酰胺染毒大鼠H反射的特征

摘  要:目的观察丙烯酰胺(ACR)不同染毒时间大鼠后肢骨间肌H反射的特征,为丙烯酰胺中毒的早期诊断提供依据。方法雄性SD大鼠64只随机分为对照组和染毒组,每组32只。再将每组随机各分为0、3、6和9 d亚组,每个亚组8只动物。染毒组腹腔注射ACR生理盐水溶液50 mg/kg(浓度5 mg/mL,容量1 mL/100 g),1次/d,对照组同种方式注射生理盐水。各组分别于注射的第0、3、6和9天测量体重及步态评分,并同步检测H反射的变化。结果与对照组比较,染毒组大鼠体重于染毒的第6天(P<0.05)和9天(P<0.01)均明显下降,步态分值在染毒的第3天(P<0.05)、6天(P<0.01)和9天(P<0.01)均高于对照组。H反射检测发现,染毒组于染毒的第3天,除H波阈值增高(P<0.05)外,H波潜伏期、H波幅度最大值(Hmax)和M波阈值与潜伏期及M波幅度最大值(Mmax)均与对照组第3天差异无统计学意义(P>0.05);染毒组于染毒的第6天,伴随着H波阈值增高(P<0.01)的同时,Hmax明显减小(P<0.05),但H波潜伏期、M波阈值和潜伏期及Mmax无明显变化(P>0.05);染毒组于染毒的第9天,H波和M波各项指标均发生了改变,表现为H波阈值(P<0.01)和M波阈值(P<0.05)均增大、Hmax(P<0.01)和Mmax(P<0.05)明显减小及H波潜伏期(P<0.05)和M波潜伏期(P<0.01)均延长。结论ACR染毒可导致H反射减弱,可能的机制为首先低阈值的Ia型粗大的有髓传入纤维的兴奋性降低,其次低阈值的Ia型传入纤维损伤,进一步损伤由α运动神经元、其所支配的骨骼肌和神经肌肉接头三者共同构成的运动单位。H反射可能是ACR中毒的一个早期诊断指标。Objective To investigate the characteristics of H-reflex of interosseous muscles of the hindlimbs by exposure to acrylamide(ACR)in rats,so as to provide the basis for the early diagnosis of acrylamide poisoning.Methods Sixty four male SD rats were randomly divided into the control group and the exposure group.The rats were intraperitoneally injected with ACR in the exposure group and the same amount of normal saline was given in the control group.Both groups were further divided into Day 0,3,6 and 9 subgroup.Body weights of rats were observed and the gait scores of rats were measured on Day 0,3,6 and 9.H-reflex of rats was determined on Day 0,3,6 and 9.Results Compared with the control group,body weights of rats were significantly reduced in the exposure group on Day 6(P<0.05)and 9(P<0.01).The gait scores on Day 3,6 and 9 were significantly higher in the exposure group than threshold(P<0.05).There was no significant difference in the latency,the maximum amplitude in H wave(Hmax),the threshold and latency of M wave,or the maximum amplitude of M wave(Mmax)between the exposure group compared and the control group on Day 3(P>0.05).In the exposure group on Day 6,the H wave threshold was significantly increased(P<0.01),and Hmax was significantly reduced(P<0.05).However,there was no significant change in the latency of H-wave,M wave or Mmax(P>0.05).The indexes of H wave and M wave were significantly changed on Day 9 of exposure in the exposure group,which showed significant increases of the thresholds of H wave(P<0.01)and M wave(P<0.05),significant reduction in Hmax(P<0.01)and Mmax(P<0.05),and significant prolonged of the latencies of H wave(P<0.05)and M wave(P<0.01).Conclusion ACR can weaken the H-reflex.The possible mechanism is that the excitability of IA type coarse myelinated afferent fibers with low threshold is reduced first,then afferent fibers with low threshold are damaged,and skeletal muscle and neuromuscular joint are further damaged.H-reflex may be an early diagnostic index of ACR poisoning.
