摘 要:目的探究核受体视黄酸X受体α(RXRA)基因在头颈部恶性肿瘤中的表达水平,分析其表达水平与临床特征的关系。方法TCGA数据库分析头颈部恶性肿瘤组织和癌旁组织中RXRA表达水平;在收集的头颈部恶性肿瘤临床组织样本中采用RT-qPCR分析RXRA的表达水平,分析其与肿瘤大小、淋巴结转移、分化程度等临床特征的关系;利用TCGA数据库分析RXRA启动子的甲基化水平,分析RXRA与DNA甲基转移酶相关基因的表达相关性;基于GEPIA工具分析DNMT1、DNMT3A和DNMT3B在头颈部恶性肿瘤中的表达水平。结果RXRA在TCGA数据库519例头颈部恶性肿瘤组织中高表达,其表达与肿瘤组织分型无显著相关性;70例头颈部恶性肿瘤组织中RXRA表达高于癌旁组织,且RXRA与肿瘤大小、淋巴结转移、组织分化程度呈正相关关系;头颈部恶性肿瘤组织中RXRA启动子的甲基化水平显著低于癌旁组织,并且RXRA表达与DNA甲基转移酶DNMT1、DNMT3A、DNMT3B表达水平呈正相关;DNA甲基转移酶DNMT1、DNMT3A和DNMT3B在头颈部恶性肿瘤组织中表达显著高于癌旁组织。结论RXRA在头颈部恶性肿瘤中高表达,且与肿瘤大小、分化程度、淋巴结转移存在正相关的联系;在头颈部恶性肿瘤中RXRA启动子的低甲基化水平是RXRA高表达的重要因素,且RXRA与DNA甲基转移酶存在紧密联系。Objective To investigate the expression of RXRA in head-neck squamous cancer(HNSC),and to analyzed the correlation between RXRA expression and clinicopathological factors.Methods The levels of RXRA was analyzed in the tumor tissues and adjacent tissues of TCGA HNSC database.RT-qPCR was used to evaluate the expression of RXRA in the collected clinical HNSC tissues,and investigate the associations between its expression and tumor size,lymph node metastasis and histologic grade.The promoter methylation level of RXRA was analyzed in TCGA HNSC tissues.The correlation level of RXRA and DNA methyltransferase was also analyzed.The expression of DNMT1,DNMT3 A and DNMT3 B was evaluated by GEPIA portal.Results RXRA was overexpressed in the 519 cases of HNSC tumor,and its expression had no significant correlation with tumor types.Also,RXRA overexpression in HNSC tumors was confirmed in a total of 70 collected clinical tissues,its level was positively correlated with tumor size,lymph node metastasis and the histologic grade.The promoter methylation level of RXRA in HNSC was also down-regulated compared with the adjacent tissues,and RXRA level was positively correlated with DNMT1,DNMT3 A and DNMT3 B.The expression of DNMT1,DNMT3 A and DNMT3 B was also highly expressed in the HNSC tumors compared with the adjacent tissues.Conclusion RXRA is overexpressed in the HNSC,and its expression has positively correlation with tumor size,histologic grade and lymph node metastasis.The low promoter methylation level of RXRA contributed to the overexpression of RXRA in HNSC,and RXRA has closely association with DNA methyltransferase.
