摘 要:本研究采用直接测序法检测了无量山乌骨鸡磷脂酰肌醇-4-磷酸3-激酶催化亚基2型α(PIK3C2A)基因内含子的单核苷酸多态性(SNPs),并分析其与体尺和屠体性状的关联性。研究发现,在鸡PIK3C2A基因内含子上共检测到5个SNPs。关联分析表明,g.38511T>A位点与龙骨长和胫围相关(P<0.05),g.38573C>T位点与盆骨宽和胫围相关(P<0.05),g.38632T>C和g.38662A>G位点与胸角和胫围相关(P<0.05),g.38729C>T位点与胸角和盆骨宽相关(P<0.05或P<0.01);g.38511T>A位点与屠体率相关(P<0.05),g.38632T>C位点与活重、屠体重、半净膛重、全净膛重、腹脂重和腹脂率相关(P<0.05或P<0.01),g.38662A>G和g.38729C>T位点与活重、屠体重、半净膛重、全净膛重和胸肌率相关(P<0.05或P<0.01)。本研究结果表明,PIK3C2A基因的5个SNPs位点与无量山乌骨鸡的体尺和屠体性状相关(P<0.01),可作为鸡体尺和屠体性状的分子标记辅助选种。In this study,single nucleotide polymorphisms(SNPs)of phosphatidylinositol-4-phosphate 3-kinase catalytic subunit type 2 alpha(PIK3C2A)gene were detected by direct sequencing in Wuliang Mountain Black-bone chickens,and its relationship with the body size and carcass traits was analyzed.Five SNPs were detected in intron region of the PIK3C2A gene.The correlation analysis showed that the g.38511T>A locus was associated with keel length and fossil bone length(P<0.05),g.38573C>T site associated with pelvic width and shank circumference(P<0.05),g.38632T>C and g.38662A>G sites associated with the breast angel and shank circumferences(P<0.05),and g.38729C>T site associated with breast angle and pelvic width(P<0.05 or P<0.01).As for the carcass traits,the g.38511T>A site was correlated with the percentage of half-eviscerated yield(P<0.05),g.38632T>C correlated with live weight,dressed weight,half-eviscerated weight,eviscerated weight,abdominal fat weight and the percentage of abdominal fat(P<0.05 or P<0.01),g.38662A>G and g.38729C>T sites associated with live weight,dressed weight,half-eviscerated weight,eviscerated weight and the percentage of breast muscle(P<0.05 or P<0.01).The above results indicated that the five SNPs of the PIK3C2A gene were correlated with the body size and carcass traits in chickens(P<0.01),and PIK3C2A gene could be the candidate gene for molecular marker-assisted selection in Wuliang Moutain black-bone chicken breeds.
