摘　　要：采用真皮间充质干细胞分泌因子治疗老化小鼠皮肤创伤,旨在阐明干细胞分泌因子对老化小鼠皮肤创伤治疗效果及其相关调控机制。体内实验采用病理组织切片,观察创面皮肤组织学变化。体外实验采用MTT assay、蛋白质免疫印迹法检测含有干细胞分泌因子的条件培养基对小鼠真皮成纤维细胞增殖的影响。结果表明,干细胞分泌因子组与对照组相比能够明显促进老化皮肤创伤愈合;干细胞分泌因子组真皮成纤维细胞存活率更高,增殖相关蛋白PCNA,Cyclin D1表达水平上升,P-ErK蛋白表达水平上升。综上所述,干细胞分泌因子能够通过ErK信号通路促进真皮成纤维细胞增殖,进而加速老化皮肤创伤愈合。研究为治疗老化皮肤创伤提供新思路,为干细胞制剂的临床应用提供理论基础。Dermal mesenchymal stem cells combined with secretory factors were used to treat the whole skin excision model of aging mice to elucidate the therapeutic effect of stem cell secretory factor on skin trauma in aged mice and its regulatory mechanism.The alter of physiological function of mouse skin tissues was measured by Histopathologic staining in vivo and the effect of conditioned medium containing stem cells compound secretory factor on the proliferation of mouse dermal fibroblasts was investigated by Western blotting and MTT assay in vitro.Our results showed that,compared with the control group,the secretory factor group significantly improved the wound healing,the survival rate of dermal fibroblasts in secretory factor group was higher,and the expression levels of proliferation-related protein PCNA and cyclin D1 increased;the expression levels of p-ErK increased.Finally,stem cell secretory factors regulated the proliferation of dermal fibroblasts through the Erk1/2 signaling pathway,and regulated the wound healing in aging skin which provided a new way to treat aging skin injury and a theoretical basis for the cliAnical application of stem cell preparation.
