摘 要:为了确定从酒泉市某养殖场北极狐(Alopex lagopus)肠道分离蛔虫的种类,本次研究通过形态学和分子生物学方法对分离的虫株进行鉴定。在对6条分离于北极狐小肠蛔虫进行形态学鉴定的基础上,应用PCR技术对分离株的线粒体cox1基因部分序列进行扩增、测序与分析。形态学鉴定结果表明6条蛔虫与弓首蛔虫形态学特征基本一致;测序结果显示,6个虫株cox1基因序列大小为418 bp,其序列同源性为99.4%—100.0%,与GenBank收录的狮弓蛔虫分离株同源性最高,为99.4%—99.7%;NJ系统进化分析结果显示6个分离株与狮弓蛔虫属于同一分支,其亲缘性较其他蛔虫种属相对最近。结合形态学与分子生物学技术将从狐狸肠道分离的蛔虫虫株均为狮弓蛔虫,其研究结果为狐狸蛔虫病的防控提供指导。We identified the species of ascaris isolated from the intestines of Alopex lagopus in a breeding farm at Jiuquan City by use of morphological and molecular biological methods.Based on the morphological identification of six isolated Ascaris spp.,the partial sequence of mitochondrial cox1 gene of the isolated strain was amplified by PCR,sequenced,and analyzed.The results of morphological identification showed that the morphological characteristics of six Ascaris strains were consistent with those of Toxascaris leonina,further results showed that the cox1 gene sequences of six strains was 418 bp,the sequence homology was 99.4%-100%,the highest homology was 99.4%-99.7%with T.leonina isolates.The constructed phylogenetic tree showed that six strains belonged to the same branch with T.leonina.Combined with morphological and molecular biological techniques,all the Ascaris strains isolated from the intestine of fox were identified as T.leonina.The results provide guidance for the prevention and control of Ascaris in fox.
