摘 要:为分析马流产沙门氏菌鞭毛蛋白FliC对牛源金黄色葡萄球菌(Staphylococcus aureus)EsxB免疫增强作用,用表达纯化的FliC与EsxB单独及联合免疫昆明小鼠,并以S.aureus分离株GW20-2攻击,检测免疫小鼠的抗体水平、攻毒保护力、体重变化、主要内脏器官荷菌数,观察肝、脾、肺及肾组织的病理组织学变化,通过real-time荧光定量PCR对免疫鼠脾细胞TLR2、TLR3、TLR4、MHC-Ⅰ、TCR的表达水平进行检测。结果表明,FliC与EsxB联合免疫后对小鼠攻击保护率可达87.5%,降低肝、脾、肺及肾组织荷菌量,且FliC与EsxB联合免疫组的TLR2、TLR3、TLR4、MHC-Ⅰ、TCR表达水平高于EsxB单独免疫组。FliC可显著增强EsxB的免疫保护效果。To investigate the immuological enhancement of Salmonella abortus equi flagellin protein(FliC)on EsxB protein of Staphylococcus aureus,in this study the expressed and purified FliC and EsxB were used to immunize Kunming mice,then the mice were challenged with S.aureus isolate GW20-2.The antibody level,titer,challenge protection,body weight change and bacterial load in liver,spleen,lung,and kidney and histopathologecial observation were investigated.In addition,the expressions of TLR2,TLR3,TLR4,MHC-Ⅰ and TCR in spleen were quantified by real-time PCR.The result indicated that FliC and EsxB vaccinated in combination could provide 87.5% challenge protection and decrease bacterial load of liver,spleen,lung and kidney.The expression level of TLR2,TLR3,TLR4,MHC-Ⅰ and TCR in FliC and EsxB immunized in combination were higher than that of EsxB immunized alone.This data demonstrated that FliC could effectively improve the immunity and protection of EsxB.
