疏肝健脾清热方对功能性消化不良大鼠胃黏膜组织5-HT1A受体及CREB蛋白表达的影响

摘　　要：目的:观察疏肝健脾清热方对功能性消化不良(FD)模型大鼠胃黏膜组织五羟色胺1A(5-HT1A)受体及环磷腺苷效应元件结合蛋白(CREB)表达的影响,探讨本方治疗FD可能的作用机制。方法:将40只SPF级Wistar大鼠随机分为空白组(N组)、模型组(M组)、西药组(WM组)、中药组(TM组),每组各10只。除N组外,其余各组大鼠采用夹尾刺激+不规则饮食的复合造模方法制备FD模型,造模成功后N组自然饲养,M组予0.9%氯化钠溶液灌胃,WM组给予莫沙必利灌胃,TM组给予疏肝健脾清热颗粒灌胃,连续治疗14d。光镜下观察各组大鼠胃窦组织病理学表现;每日观察大鼠一般情况并记录大鼠体质量变化;免疫荧光法检测大鼠胃窦5-HT1A受体含量,Western Blot法检测大鼠胃CREB蛋白表达。结果:光镜下观察各组大鼠胃窦组织病理学变化未见明显异常,疏肝健脾清热方能有效改善FD模型大鼠一般情况,增加FD大鼠体质量;降低FD大鼠胃黏膜组织5-HT1A受体含量,增加其CREB蛋白表达。结论:疏肝健脾清热方能降低胃黏膜组织5-HT1A的含量、增加CREB蛋白表达,这可能是本方治疗FD的作用机制之一。Objective:To observe the effects of Shugan Jianpi Qingre Formula on the expression of 5-HT1A receptor and CREB protein in gastric mucosa of FD rats.Methods:Forty SPF grade Wistar rats were randomly divided into a blank group(N group),model group(M group),western medicine group(WM group)and traditional Chinese medicine group(TM group),10 in each group.After the successful model-making,the rats in the N group were fed naturally,the WM group was given mosapride,the TM group was given Shugan Jianpi Qingre Formula,and the rats were treated for 14 d.The histopathological features of each group were observed under light microscope,the general condition of rats was observed daily and the body weight of rats was recorded,the content of 5-HT1A receptor in rat antrum was detected by immunofluorescence,and the expression of CREB protein in rat stomach was detected by Western Blot.Results:The histopathological changes of gastric antrum of rats were not abnormal under light microscope.Shugan Jianpi Qingre Formula could effectively improve the general situation of FD rats and increase the body weight of FD rats.Conclusion:Shugan Jianpi Qingre Formula can reduce the content of 5-HT1A in gastric mucosa and increase the expression of CREB protein,which may be one of the mechanisms of treating FD.
