摘  要:目的:观察香苓开胃方对功能性消化不良大鼠(FD)干预效应及机制。方法:测定FD大鼠肠推进率,酶联免疫吸附法(ELISA)检测血清胃动素(MTL)、胃饥饿素(Ghrelin)和胃泌素(GAS)水平,实时定量PCR检测与分析瘦素(Leptin)、闭合蛋白(Occludin)、络氨酸激酶(C-kit)mRNA表达。结果:香苓开胃方高剂量药物干预显著促进肝郁脾虚模型FD大鼠肠推进率(P<0.05);显著提高大鼠血清MTL(P<0.05);显著下调FD大鼠胃组织Leptin mRNA表达(P<0.05)和提高FD大鼠胃组织Occludin mRNA表达(P<0.05)。结论:香苓开胃方对FD具有显著的治疗作用,其机制与促进胃肠运动有关、调节胃肠激素等有关。Objective:To observe the effects and mechanism of Xiangling Kaiwei Formula on model of functional dyspepsia(FD)in rats.Methods:The FD rats intestinal propulsive rate was determined.The levels of serum motilin(MTL),Ghrelin and gastrin(GAS)were detected by enzyme-linked immunosorbent assay(ELISA).The expression of Leptin,Occludin and tyrosine kinase(c-kit)mRNA were detected and analyzed by real-time quantitative PCR.Results:High-dose of Xiangling Kaiwei Formula significantly improved intestinal propulsion rate(P<0.05),significantly increased theserum MTL(P<0.05),significantly decreased the expression of leptin mRNA in gastric tissue of FD rats(P<0.05),and increased the expression of occludin mRNA in gastric tissue of FD rats(P<0.05).Conclusion:Xiangling Kaiwei Formula has a significant therapeutic effect on functional dyspepsia.The mechanism is related to promoting gastrointestinal movement and regulating gastrointestinal hormones.
