摘 要:目的:观察赪桐根3种提取部位对炎症组织及血清中主要炎症因子表达的影响。方法:建立去肾上腺小鼠、佐剂性关节炎大鼠模型,采用ELISA法测定小鼠致炎组织液中丙二醛(MDA)、一氧化氮(NO)、超氧化物歧化酶(SOD)及大鼠血清中肿瘤坏死因子-α(TNF-α)、前列腺素E2(PGE2)、白介素-1β(IL-1β)、白介素-6(IL-6)、白三烯B4(LTB4)的含量。结果:与模型组比较,正丁醇、95%乙醇部位对去双侧肾上腺小鼠足肿胀有明显抑制作用;正丁醇部位对双侧肾上腺小鼠耳肿胀有明显抑制作用;3个部位均能降低致炎足组织液中MDA、NO含量并提高SOD活性,可显著降低佐剂性关节炎大鼠血清中TNF-α、PGE2、IL-1β、IL-6、LTB4的含量。结论:赪桐根3个部位抗炎作用可能部分依赖于下丘脑-垂体-肾上腺(HPAA)系统,并且与调节MDA、NO、TNF-α、PGE2、IL-1β、IL-6、LTB4的合成与分泌,提高SOD活性有关。Objective:To observe the effect of three extracts of roots of Clerodendrum japonicum on the expression of main inflammatory factors in inflammatory tissues and serum.Methods:The models of bilateral adrenalectomy mice and djuvantinduced arthritis rats were established.ELISA was used to detect the contents of MDA,NO,SOD in inflammatory tissue fluid of mice and TNF-α,IL-1β,IL-6,PGE2,LTB4 in serum of rats.Results:Compared with the model group,N-butanol extract and 95%ethanol extract had significant inhibitory effects on the swelling of the feet of the bilateral adrenalectomy mice;N-butanol extract had significant inhibitory effects on the swelling of the ears of the bilateral adrenalectomy mice;the three extracts could reduce the MDA and NO contents in the inflammatory foot tissue fluid and increase the activity of SOD,and could significantly reduce the TNF-α,PGE2,IL-1β,IL-6 and LTB4 contents in the serum of the adjuvant-induced arthritis rats.Conclusion:The antiinflammatory effect of the three extracts of roots of Clerodendrum japonicum may be partly dependent on the HPAA system,and is related to the regulation of the synthesis and secretion of MDA,NO,TNF-α,PGE2,IL-1β,IL-6,LTB4 and the enhancement of activity of SOD.
