指针法对胰岛素抵抗大鼠骨骼肌Sirt1/PGC-1α信号通路的影响

摘　　要：目的:探讨指针法治疗对胰岛素抵抗(IR)大鼠骨骼肌Sirt1/PGC-1α信号通路的影响。方法:SPF级雄性SD大鼠75只随机选取10只为正常组,其余65只建立IR模型。模型组按35mg/kg注射STZ溶液,空白组注射等体积的柠檬酸-柠檬酸钠缓冲液。将造模成功的50只IR大鼠,随机重新分为模型组、对照组(白藜芦醇)、指针1组(低刺激量)、指针2组(中刺激量)和指针3组(高刺激量),各10只。每日1次,均治疗4周。结果:与模型组比较,对照组、指针2组、指针3组AMPK、Sirt1、PGC-1α蛋白表达水平,AMPK、Sirt1/PGC-1αmRNA表达水平均显著提高(P<0.01);其中指针2组(中刺激量)疗效具有显著差异(P<0.01)。结论:指针法治疗胰岛素抵抗具有一定的优势,不同时长指针法操作疗效存在差异,其中指针2组(10min)疗效与对照组相当,作用机制可能和上调骨骼肌Sirt1/PGC-1α信号通路相关基因和蛋白表达水平有关。Objective:To explore the effects of Zhizhen therapy on Sirt1/PGC-1αsignaling pathway in rats with insulin resistance(IR).Methods:Ten of seventy-five SPF-grade male rats were randomly selected into a normal group,and the remaining 65 rats were used to establish insulin resistance animal models.The model group was injected with STZ solution at 35 mg/kg,and the control group was injected equal volume of citric acid-sodium citrate buffer to establish insulin resistance animal models.Fifty IR model rats were randomly divided into model group,control group(resveratrol),Zhizhen group 1(low stimulation),Zhizhen group 2(medium stimulation)and Zhizhen group 3(high stimulation),each group included 10 rats.Treating once a day for 4 weeks.Results:Compared with model group,the expression levels of AMPK,Sirt1 and PGC-1αin control group,Zhizhen 2 group and Zhizhen 3 group,and the expression levels of AMPK and Sirt1/PGC-1αgene were significantly increased(P<0.01),and the curative effect of Zhizhen 2 group was significantly different(P<0.01).Conclusion:Zhizhen therapy has certain advantage for the treatment of insulin resistance.The therapeutic effect of different length of Zhizhen therapy was different,among which the therapeutic effect of Zhizhen 2 group(10 min)was similar to that of control group.The possible mechanism may be related to the up-regulation of genes and protein expression levels in Sirt1/PGC-1αsignaling pathway.
