摘  要:目的:研究青娥方对去卵巢大鼠骨组织Src/p130Cas/CRK信号通路的影响。方法:96只3月龄雌性SD大鼠,随机分为假手术组、单纯去卵巢组、骨疏康组(去卵巢+骨疏康颗粒组)、青娥方组(去卵巢+青娥方组)。前两组灌服0.9%氯化钠溶液,后两组分别灌服骨疏康及青娥方。分别于用药4周、8周、12周检测大鼠腰椎Src/p130Cas/CRK mRNA及蛋白含量、右股骨颈骨密度(BMD)、右股骨最大载荷、左股骨颈骨小梁面积百分比(BV/TV)等指标的变化。结果:去势后4周,单纯去卵巢组Src mRNA、CRK mRNA及蛋白表达升高,在8、12周有所下降,而p130Cas mRNA持续升高,8周达到峰值,均显著高于同期假手术组(P<0.01,P<0.05)。股骨颈骨、股骨最大载荷及股骨颈骨小梁面积百分比随着时间推移逐渐降低。青娥方干预4、8、12周后,Src mRNA、p130Cas mRNA、CRK mRNA及蛋白表达持续降低,低于单纯去卵巢组(P<0.01,P<0.05)。股骨颈BMD在用药后4周有所下降,在用药后8、12周BMD显著高于单纯去卵巢组(P<0.01,P<0.05)。而股骨最大载荷持续升高,在8、12周显著高于单纯去卵巢组(P<0.05,P<0.01),但仍低于假手术组。股骨颈骨小梁面积百分比持续下降,下降速度随时间推移明显减缓,在12周显著高于单纯去卵巢组(P<0.01,P<0.05)。结论:青娥方能抑制骨组织Src/p130Cas/CRK的表达,提高骨密度、股骨最大载荷及股骨颈骨小梁面积百分比,降低去卵巢大鼠的骨吸收速度。Objective:To study the effects of Qing’e Formula on Src/p130 Cas/CRK signaling pathway in bone tissue of ovariectomized rats.Methods:Ninty-six 3-month-old female SD rats were randomly divided into sham operation group(sham group),ovariectomy group(OVX group),Gushukang group(ovariectomized+Gushukang granule group)and Qing’e Formula group(ovariectomized+Qing’e Formula group).Rats in the former two groups were gavaged administration with normal saline,and rats in Gushukang group and Qing’e Formula group were gavaged with Gushukang granule and with Qing’e Formula,respectively.The mRNA and protein content of Src/p130 Cas/CRK in lumbar vertebra,bone mineral density(BMD)of right femoral neck,maximum load of right femur and percentage of trabecular area(BV/TV)of left femoral neck were detected in the 4 th,8 th and 12 th weeks after treatment.Results:The mRNA and protein expression of Src and CRK in the rats of OVX groups increased significantly in 4 th weeks and declined in 8 th and 12 th weeks,while p130 cas mRNA continued to increase and reached the peak at 8 weeks,which were significantly higher than those in sham operation group at the same period(P<0.01,P<0.05).The BMD of femoral neck,the maximum load of femur and BV/TV of femoral neck gradually decreased with time.After treating for 4,8 and 12 weeks with Qing’e Formula,the mRNA and protein expression of Src,p130 Cas and CRK in bone tissue decreased continuously,and it was significantly lower than that of OVX group(P<0.01,P<0.05).It was even lower than that of sham operation group at the same period.BMD of femoral neck decreased in 4 th week after treatment,and was significantly higher than that of OVX group at 8 th and 12 th weeks after treatment(P<0.01,P<0.05).The maximum load of femur increased in 8 th and 12 th weeks,which was significantly higher than that of OVX group(P<0.01,P<0.05),but still lower than that of sham operation group.The BV/TV of femoral neck decreased,and the rate of decline slowed down with the passage of time.At 12 weeks,it was si
