摘 要:目的:基于腺苷酸活化蛋白激酶(AMPK)及其基因的表达水平,探讨加味温胆汤有效改善雌性营养性肥胖大鼠脂质代谢紊乱的作用机制和量效关系。方法:将50只SD雌性大鼠按体质量随机分为5组,即正常对照组,模型对照组,加味温胆汤高、中、低剂量组(18.2、9.1、4.55g/kg)。除正常对照组外,其余均采用"普通饲料+高脂乳剂+碳酸饮料"混合饲养法建立肥胖大鼠模型。经末次给药后,麻醉取材,采用酶联免疫吸附测定分别检测肝、腓肠肌组织中AMPKβ的表达;采用实时荧光定量PCR分别检测肝AMPKβmRNA、脂肪AMPKα1mRNA、腓肠肌AMPKα2 mRNA的表达水平。结果:与模型对照组比较,加味温胆汤高剂量组显著下调腓肠肌AMPKα2 mRNA的表达(P<0.05),加味温胆汤中剂量组显著下调脂肪AMPKα1 mRNA、腓肠肌AMPKα2 mRNA的表达(P<0.05);加味温胆汤低剂量组显著下调肝AMPKβ、脂肪AMPKα1 mRNA和腓肠肌AMPKα2 mRNA的表达(P<0.05);与加味温胆汤高剂量组比较,加味温胆汤中、低剂量组显著下调大鼠脂肪AMPKα1 mRNA的表达(P<0.05)。结论:加味温胆汤抗雌性大鼠营养性肥胖的作用机制与下调大鼠肝和腓肠肌组织中AMPK及其基因的表达水平有关。Objective:Based on the expression level of amp-activated protein kinase(AMPK)and its gene,the mechanism and dose-effect relationship of Jiawei Wendan Decoction in improving lipid metabolism disorder in female nutritional obese rats were studied.Methods:Fifty SD female rats were randomly divided into 5 groups according to body weight.namely normal control group,model control group,high,medium and low dose group of Jiawei Wendan Decoction(18.2,9.1,4.55 g/kg).In addition to the normal control group,the other rats were fed with‘common feed+high fat emulsion+carbonated beverage’to establish the model of nutritional obesity,after the last drug administration for animals in each group,the rats were anaesthesia to gain material.the levels of AMPK β in liver and gastrocnemius muscle tissues were detected by Elisa.the expression levels of liver tissue AMPKβmRNA,adipose tissue AMPKα1 mRNA and AMPKα2 mRNA in gastrocnemius muscle were detected by RT-PCR.Results:Compared with the model group,the high-dose group of Jiawei Wendan Decoction could significantly downregulate the expression of AMPKα2 mRNA in the gastrocnemius muscle(P<0.05).The middle dose group can significantly down-regulate the expression of AMPKα1 mRNA in adipose tissue and AMPKα2 mRNA in gastrocnemius muscle(P<0.05).the low-dose group significantly down-regulated the expression levels of liver tissue AMPKβ,adipose tissue AMPKα1 mRNA and AMPKα2 mRNA in gastrocnemius muscle(P<0.05).Compared with the high-dose group of Jiawei Wendan Decoction,the expression levels of AMPKα1 mRNA in the middle and low dose groups were significantly lower than those in the high dose group(P<0.05).Conclusion:The mechanism of Jiawei Wendan Decoction against nutritional obesity in female rats is related to the regulation of AMPK expression in liver,gastrocnemius and adipose tissue.
