复方生/炒白芥子穴位贴敷对变应性鼻炎大鼠行为学及血清IgE、IL-4、IFN-γ的影响

摘　　要：目的:观察生/炒白芥子穴位贴敷对变应性鼻炎大鼠行为学及血清免疫球蛋白E(IgE)、白细胞介素4(IL-4)、γ干扰素(IFN-γ)的影响,探讨不同炮制药物穴位贴敷的疗效及干预变应性鼻炎的作用机制。方法:Wistar大鼠随机分为对照组、模型组、丙酸氟替卡松组、生白芥子组、炒白芥子组,每组10只,以卵白蛋白致敏激发法建立变应性鼻炎模型。造模后,模型组予0.9%氯化钠溶液滴鼻,每侧鼻孔5μL,每日1次,共14次;丙酸氟替卡松组予丙酸氟替卡松滴鼻,方法同模型组;生白芥子组予穴位贴敷干预,取双侧"肺俞""脾俞""肾俞",贴敷2h,隔日1次,共贴7次;炒白芥子组选用炒白芥子贴膏,方法同生白芥子组。检测大鼠造模前、造模后及治疗后行为学变化。采用酶联免疫吸附法检测治疗后大鼠血清中IgE、IL-4、IFN-γ含量。结果:与模型组比较,经治疗后生白芥子组及炒白芥子组行为学评分和IgE、IL-4含量显著降低(P<0.05),IFN-γ含量显著升高(P<0.05),且与丙酸氟替卡松组无显著性差异。而生白芥子组在行为学评分及IFN-γ含量改善方面均优于炒白芥子组(P<0.05)。结论:穴位贴敷干预可有效改善变应性鼻炎大鼠行为学,其作用机制可能与通过下调血清IL-4、上调血清IFN-γ水平来调节Th1/Th2平衡,及进一步下调IgE有关,穴位贴敷组方中选用生白芥子疗效更佳。Objective:To observe the effects of acupoint application of raw/fried Semen sinapis on the behavior and serum immunoglobulin E(IgE),interleukin-4(IL-4)and interferon-gamma(IFN-γ)in rats with allergic rhinitis.And to discuss the curative effect of acupoint application of different processed drugs and the mechanism of intervening allergic rhinitis.Methods:Wistar rats were randomly divided into control group,model group,fluticasone propionate group,raw semen sinapis group and fried semen sinapis group,10 rats in each group.The allergic rhinitis model was established by ovalbumin sensitization.After modeling,the model group was given saline nasal drops,5μL of each nostril,once a day for 14 times;fluticasone propionate group was given fluticasone propionate for nose dropping,the way was the same as model group;raw Semen sinapis group was given acupoint application intervention,with bilateral‘Feishu(BL 13)’,‘Pishu(BL 20)’,‘Shenshu(BL 23)’,pasting for 2 h,once every other day,a total of 7 times;fried semen sinapis group used fried semen sinapis,the way was the same as raw semen sinapis group.Behavioral changes of base,post-modeling and post-treatment were examined.The serum levels of IgE,IL-4 and IFN-γ were detected by enzyme-linked immunosorbent assay.Results:Compared with the model group,the behavioral scores,IgE and IL-4 content in the raw&fried semen sinapis group were significantly decreased(P<0.05),and the IFN-γcontent was significantly increased(P<0.05).There was no significant difference with the fluticasone propionate group.The behavioral scores and the IFN-γ content of the raw semen sinapis group were significantly higher than those of the fried Semen sinapis group(P<0.05).Conclusion:Acupoint application can effectively improve the behavior of rats with allergic rhinitis.The mechanism may be related to the regulation of Th1/Th2 balance by down-regulating serum IL-4,up-regulating serum IFN-γ level,and further down-regulating IgE.The selection of raw semen sinapis was better in the acupoint app
