摘 要:目的:研究复方茵丹汤对α-萘异硫氰酸酯(ANIT)诱导的大鼠急性肝内胆汁淤积(AIC)作用机制。方法:将108只雄性SD大鼠随机分为正常组,模型组,中药高、中、低剂量组和熊去氧胆酸胶囊(UDCA)组,每组18只,依据取材时间不同又将各组随机分为24h、48h、72h三个亚组,每组6只。模型组、中药组、UDCA组大鼠一次性给予2%ANIT按100mg/kg灌胃造模,正常组予等容积色拉油灌胃。造模2h后,中药高、中、低剂量组分别给予生药含量为24.48、12.24、6.12g·kg^-1·d^-1的复方茵丹汤中药煎剂1mL/100g灌胃,模型组和正常组给予等容积0.9%氯化钠溶液灌胃,UDCA组给予67.5mg·kg^-1·d^-1 UDCA水溶液1mL/100g灌胃,均1次/d。造模后分别于24h、48h、72h处死相应时相大鼠,采用RT-q-PCR、Western Blot和IHC技术检测PXR及CAR在大鼠肝组织的表达水平。结果:与同时相正常组比较,模型组PXR、CAR mRNA及蛋白表达显著降低(P<0.05,P<0.01)。与同时相模型组比较,中药各剂量组及UDCA组PXR、CAR mRNA及蛋白表达均显著升高(P<0.05,P<0.01)。中药高剂量组PXR、CAR基因及蛋白表达显著高于中药低剂量组和UDCA组(P<0.05,P<0.01)。结论:复方茵丹汤对ANIT灌胃诱导AIC大鼠有防治作用,可能与其上调PXR、CAR mRNA和蛋白表达有关。Objective:To investigate the mechanism of compound Yindan Decoction on acute intrahepatic cholestasis induced by α-naphthalene isothiocyanate(ANIT)in rats.Methods:A total of 108 male SD rats were randomly divided into normal control group,model group,traditional Chinese medicine(TCM)high,medium and low dose group and positive control group,18 rats in each group.According to the different processing time,each group is divided into 24-hour,48-hour,72-hour three sub-groups in turn,each group had 6 rats.The rats of model group,TCM groups and positive control group were given separately 2%ANIT and equal volume of salad oil was given to the normal group once administration by gavage.After 2 hours of modeling,high,medium and low dose groups were treated with service compound Yindan Decoction of traditional Chinese medicine preparation 24.48,12.24,6.12 g·kg^-1·d^-1,model group and normal control group received such as volume of physiological saline gavage,positive drug control group was treated with 67.5 mg·kg^-1·d^-1 ursodeoxycholic acid aqueous solution orally,1 time/d.24 h,48 h and 72 h were sacrificed at the corresponding time of the model,and the liver function indexes of rats in each group were dynamically observed by enzyme linked immunosorbent assay,and the pathological changes of liver tissue dealt with hematoxylin-eosin staining were observed by microscope.Real-time PCR,Western Blot and immunohistochemistry(IHC)were used to detect the expression of PXR and CAR in rat liver.Results:Compared with the simultaneous phase normal group,the expression of PXR and CAR mRNA and proteins in the model group were significantly decreased(P<0.05,P<0.01).Compared with the simultaneous phase model group,the expressions of PXR and CAR mRNA and proteins in each dose group and ursodeoxycholic acid group were significantly increased(P<0.05,P<0.01).The expression of PXR and CAR mRNA and proteins in the high-dosage TCM group was higher than that in the simultaneous phase low dose group and ursodeoxycholic acid group(P<0.05,P<0.0
