摘  要:目的:研究百合地黄汤合酸枣仁汤对抑郁症模型大鼠海马脑源性神经营养因子(BDNF)和血清肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-6、IL-1表达的影响及其抗抑郁的作用机制。方法:采用慢性应激结合隔离孤养的方法制造大鼠抑郁症模型,随机分为空白组、模型组、氟西汀组、百合地黄汤合酸枣仁汤(合方)高、中、低剂量组。造模与给药同时进行。空白组、模型组灌胃0.9%氯化钠溶液,各给药组灌胃相应药物,连续21d。观察记录大鼠的一般情况和体质量;"敞箱实验"对大鼠进行行为学检测;ELISA法测定大鼠血清中的TNF-α、IL-6、IL-1的含量;HE法观察大鼠脑海马病理组织形态学改变;Western Blot法测脑海马BDNF的含量。结果:与空白组比较,模型组体质量增长速度减缓、敞箱实验得分显著减少、血清TNF-α、IL-6、IL-1含量升高、脑海马BDNF含量下降(P<0.01);与模型组比较,合方高剂量组、氟西汀组大鼠的体质量增长、敞箱实验得分增加、血清TNF-α、IL-6、IL-1含量降低、脑海马BDNF含量增加(P<0.01);与氟西汀组比较,合方高剂量组体质量增长、敞箱实验得分增加、血清TNF-α、IL-6、IL-1含量降低,脑海马BDNF含量增加(P<0.05,P<0.01)。结论:百合地黄汤合酸枣仁汤通过调整血清TNF-α、IL-6、IL-1的过度分泌以及脑海马BDNF的含量,进而改善大鼠的行为学表现,这可能是百合地黄汤合酸枣仁汤抗抑郁作用机制之一。Objective:To study the effects of Baihe Dihuang Decoction and Suanzaoren Decoction on BDNF in hippocampus and serum TNF-α,IL-6,IL-1 in rats with depression and its antidepressant mechanism.Methods:The method of isolating binding lone chronic stress rat model of depression raised manufacture,were randomly divided into control group,model group,fluoxetine group,Baihe Dihuang Decoction and Suanzaoren Decoction(combined formula)high,medium and low dose group.Modeling and administered simultaneously.Control group,model group were given physiological saline-administered group each week administration,continuous 21 d.General observations and body weight of rats;‘open field experiment’behavioral tests on rats;measurement of TNF-αin rat serum by ELISA,IL-1 and IL-6 content;HE was observed in rats morphological pathological changes hippocampus;Western Blot as measured content of BDNF in hippocampus.Results:Compared with the control group,model group slowed weight gain,reduced open field test scores,increased serum TNF-α,IL-6,IL-1 content,decreased brain BDNF in hippocampus(P<0.01);Compared with the model group,weight gained,open field test scores increased,serum TNF-α,IL-6,IL-1 decreased,brain BDNF in hippocampus increased in the combined formula high dose group and fluoxetine group(P<0.01);Compared with the fluoxetine group,weight gained,open field test scores increased,serum TNF-α,IL-6,IL-1 decreased,brain BDNF in hippocampus increased in the combined formula high dose group(P<0.05,P<0.01).Conclusion:By adjusting the excessive secretion of TNF-α,IL-6 and IL-1 in serum and the content of BDNF in the hippocampus,regulating behavior of rats,it may be one of the antidepressant mechanisms of Baihe Dihuang Decoction and Suanzaoren Decoction.
