摘 要:目的:研究加味百合地黄汤对干燥综合征模型NOD小鼠血清IFN-γ以及焦虑抑郁行为的影响。方法:将NOD/Ltj小鼠随机分为模型组、对照组、中药组,C57BL/6J小鼠为正常组,正常组、模型组灌服蒸馏水,对照组灌服白芍总苷,中药组灌服加味百合地黄汤。通过检测旷场实验、高架十字迷宫实验评估小鼠行为学,对颌下腺进行HE染色,酶联免疫法检测血清IFN-γ表达水平。结果:正常组组织学评分低于各组NOD小鼠(P<0.01),各组NOD小鼠组间组织学评分无显著性差异,旷场实验显示正常组小鼠跨格次数低于模型组及对照组(P<0.01),中药组小鼠跨格次数低于对照组(P<0.01),高架十字迷宫实验显示正常组小鼠OE%、OT%低于各组NOD小鼠(P<0.05,P<0.01);对照组、中药组血清IFN-γ水平均低于模型组(P<0.01)。结论:加味百合地黄汤可下调NOD小鼠血清IFN-γ表达水平,下调NOD小鼠的自主活动性。Objective: To observe the effects of modified Baihe Dihuang Decoction on the expression of interferon-γ(IFN-γ) in serum and behaviors of anxiety and depression in the Sjogren’s syndrome(SS) non-obese diabetic(NOD) mice. Methods: C57 BL/6 J mice were used as normal group, and NOD/Ltj mice were randomly divided into model group, control group and Chinese medicine group. The normal group and the model group were given distilled water. The control group was given total glucosides of paeony. The Chinese medicine group was given modified Baihe Dihuang Decoction. The submandibular gland tissues were stained by HE, and observed under the microscope. Serum IFN-γ levels were detected by ELISA. Mice behavior is detected by the open field test(OFT) and the elevated plus-maze. Results: The histological scores of the normal group were lower than those of the NOD mice(P<0.01), and there was no significant difference in the histological scores between the groups of NOD mice. The normal group had a significantly greater number of line crossings than the model group and the control group in the open field test(P<0.01), and the Chinese medicine group had a fewer number of line crossings than the control group(P<0.01). Compared with all NOD mice group, the values of OT% and OE% obviously decreased in the normal group in the elevated plusmaze(P<0.05, P<0.01). The serum IFN-γ level of the control group and the Chinese medicine group were lower than that of the model group(P<0.01). Conclusion: Modified Baihe Dihuang Decoction can down-regulate the expression level of serum IFN-γ in NOD mice and down-regulate the autonomic activity of NOD mice.
