摘 要:目的:探讨补肾壮骨颗粒对SAMP6小鼠血清生长激素(GH)浓度的影响。方法:实验动物分为5组:SAMR1小鼠0.9%氯化钠溶液灌胃组(R1组),SAMP6小鼠分为0.9%氯化钠溶液灌胃组(P6空组)、皮下注射rhGH组(rhGH组)和补肾壮骨颗粒灌胃组(补肾组)及联合皮下注射rhGH组和补肾壮骨颗粒灌胃组(rhGH+补肾组),每组10只,每日干预1次。分别干预3、6个月后进行股骨、脊柱骨密度(BMD)检测,血清GH、骨钙素(BGP)、甲状旁腺激素(PTH)浓度测定。结果:干预3个月后:补肾组及补肾+rhGH组中股骨BMD高于P6空组及rhGH组(P<0.05)。干预6个月后:3种药物干预组中股骨BMD均比P6空组高(P<0.05);补肾组及补肾+rhGH组脊柱BMD高于P6空组(P<0.05)。两阶段干预后,与P6空组比较,3种药物干预组血清GH及BGP浓度上升、PTH浓度下降(P<0.05)。6个月干预组P6小鼠血清GH浓度与股骨(r=0.599,P<0.01)及脊柱BMD呈正相关(r=0.610,P<0.01)。结论:补肾壮骨颗粒可以提高SAMP6小鼠脊柱及股骨BMD,脊柱及股骨BMD的提高可能与血清GH浓度升高有关。Objective:To study the changes in serum growth hormone(GH)in senile osteoporosis mice after Bushen Zhuanggu Granules therapy.Methods:The experiment mice were divided into five groups:SAMR1 mice which feed saline lavage,SAMP6 mice were divided as saline lavage group,subcutaneous injection of rhGH group,Bushen Zhuanggu Granules treatment group,and subcutaneous injection of rhGH combined with Bushen Zhuanggu Granules treatment group,10 mice in each group,all intervention was conducted once a day.After intervention for 3 months and 6 months,the bone mineral density(BMD)of femur and spine,the level of serum GH,bone growth protein(BGP)and parathyroid hormone(PTH)were measured for comparison.Results:After 3 months of intervention,the BMD of femoral in tonifying kidney group and tonifying kidney combined with rhGH group was higher than that in P6 control group and rhGH group(P<0.05).After 6 months of intervention,compared to the P6 control group,the BMD of femoral in the three drug intervention groups was increase(P<0.05),the BMD of spine in tonifying kidney group and kidney tonifying combined with rhGH group increased compared with P6 control group(P<0.05).After 3 months and 6 months of intervention,compared with P6 control group,the serum GH and BGP concentrations in rhGH group,tonifying kidney group and tonifying kidney combined with rhGH group increased and PTH concentration decreased(P<0.05).Serum GH concentration in P6 mice in the 6 month intervention group was positively correlated with femur(r=0.599,P<0.01)and spine BMD(r=0.610,P<0.01).Conclusion:Bushen Zhuanggu Granules can improve the BMD of spine and femur in SAMP6 mice,the increase of BMD of spine and femur may be related to the increase of serum GH concentration.
