摘 要:目的:建立一测多评法测定皮肤康洗液中7种活性成分含量。方法:采用HPLC法,流动相为乙腈(A)-0.1%磷酸溶液(B)梯度洗脱,流速为1.0 ml·min^-1,检测波长为230 nm(芍药苷)、327 nm(绿原酸)、237 nm(甘草酸铵、甘草苷)、254 nm(大黄素、大黄酸)、322nm(蛇床子素),柱温为30℃。以芍药苷为内参物,建立一测多评法同时测定绿原酸、甘草苷、甘草酸铵、大黄酸、大黄素和蛇床子素6种成分的相对较正因子(RCF)并计算其含量,并与外标法分析比较以上7种成分的含量差异。结果:绿原酸、甘草苷、甘草酸铵、大黄酸、大黄素、蛇床子素和芍药苷分别在3.054~54.972μg·ml^-1,1.408~25.335μg·ml^-1,1.529~27.513μg·ml^-1,1.077~19.386μg·ml^-1,0.921~16.578μg·ml^-1,1.326~23.859μg·ml^-1,2.750~49.500μg·ml^-1范围内线性关系良好(r≥0.9993);平均加样回收率(RSD)分别为100.7%(1.19%)、98.8%(1.57%)、99.5%(0.99%)、100.6%(1.54%)、99.8%(1.70%)、100.5%(1.39%)、99.4%(0.93%)(n=6);一测多评法测得的计算值与外标法测定值的差异无统计学意义(P>0.05)。结论:一测多评法可用于皮肤康洗液中7种有效成分的含量测定,且方法简单、有效、结果准确。Objective:To develop a method of quantitative analysis of multi-components by single marker for Pifukang lotions.Methods:Using an HPLC method,the mobile phase was acetonitrile-0.1%physcion acid in a gradient elution mode,the flow rate was 1.0 ml·min^-1,the detection wavelengths were set at 230 nm for paeoniflorin,327 nm for chlorogenic acid,237 nm for ammonium glycyrrhizinate and liquiritin,254 nm for edomin and rhein,and 322nm for osthole,and the column temperature was 30℃.Using paeoniflorin as the internal standard,the relative correction factors of chlorogenic acid,ammonium glycyrrhizinate,liquiritin,edomin,rhein and osthole were detected by QAMS and their contents were calculated,and then the content differences of above seven components were compared with those of the external standard method.Results:The standard curves of chlorogenic acid,liquiritin,ammonium glycyrrhizinate,rhein,edomin,osthole,and paeoniflorin all showed good linearity within 3.054-54.972μg·ml^-1,1.408-25.335μg·ml^-1,1.529-27.513μg·ml-1,1.077-19.386μg·ml-1,0.921-16.578μg·ml-1,1.326-23.859μg·ml^-1,and 2.750-49.500μg·ml^-1,respectively(r≥0.9993).The average recovery was 100.7%(1.19%),98.8%(1.57%),99.5%(0.99%),100.6%(1.54%),99.8%(1.70%),100.5%(1.39),99.4%(0.93%)(n=6),respectively.There were no significant differences between the calculated values from QAMS and the estimated values from the external standard method(P>0.05).Conclusion:The QAMS method can be used for the determination of seven active constituents in Pifukang lotions,and the method is simple,effective and accurate.
