摘　　要：目的研究解毒祛瘀方对动脉粥样硬化斑块的影响机制。方法收集小鼠腹腔巨噬细胞进行体外培养,使用80 μg/mL Ox-LDL诱导巨噬细胞凋亡,以50、100、200 μg/mL解毒祛瘀方作为低、中、高3种剂量进行干预,AnnexinV-FITC和PI双染后用流式细胞仪检测细胞凋亡。将ApoE-/-小鼠随机分成6组:正常组、模型组、低浓度治疗组、中浓度治疗组、高浓度治疗组和辛伐他汀组(Simvastatin,SV),分别予生理盐水、解毒祛瘀方和辛伐他汀,每日1次灌胃,连续给药8周,收集血液并离心,检测血清中总胆固醇、三酰甘油等生化指标。结果与正常组比较,模型组细胞凋亡率明显增高(P<0.05);低、中、高剂量治疗组与模型组细胞凋亡率比较,差异均有统计学意义(P<0.05)。高脂饮食喂养组小鼠的血清总胆固醇(TC)和三酰甘油(TG)含量与正常组比较明显上升(P<0.05);治疗组与模型组TC、TG含量比较,差异有统计学意义(P<0.05)。结论解毒祛瘀方可以有效降低与AS形成密切相关的ApoE-/-小鼠的血清TC和TG水平,且抑制了巨噬细胞凋亡,有助于改善AS的进展。Objective To study the mechanism of Jiedu Quyu decoction on arteriosclerosis plaque.Methods The mouse peritoneal macrophages were collected and cultured in vitro.The apoptosis of macrophages was induced by 80 μg/mL Ox-LDL.The low,medium and high doses of 50,100,200 μg/mL Jiedu Quyu decoction were used to intervene the apoptosis,aNNEXIN V-FITC and Pi Double staining were used to detect apoptosis with Flow cytometry.ApoE-/-mice were randomly divided into 6 groups:Normal Group,model group,low concentration treatment group,medium concentration treatment group,high concentration treatment group and Simvastatin (SV) group,the blood samples were collected and centrifuged to detect the serum total cholesterol,triglyceride and other biochemical indexes.Results Compared with the normal group,the apoptosis rate in the model group was significantly higher (P<0.05),and that in the low,middle and high Dose Treatment Group was significantly higher than that in the model group (P<0.05).2.Compared with the normal diet group,TC and triglyceride in the high fat diet group were significantly higher than those in the normal diet group (P<0.05) The TC and TG contents in the treatment group were significantly higher than those in the model group (P<0.05).Conclusion Jiedu Quyu decoction can effectively reduce the serum TC and TG levels of Apo e-/-mice,which are closely related to the formation of AS,and inhibit the apoptosis of macrophages,which is helpful to improve the progress of AS.
