摘 要:【目的】应用乳腺癌细胞(SK-BR-3)研究3种还原性中药成分与巯基丙酸修饰的碲化镉(MPA-CdTe)量子点的联合作用,并研究相关机制。【方法】分别以甘草酸、水飞蓟宾及熊去氧胆酸作为中药成分代表,考察其与量子点联用情况下对量子点毒性的减弱作用,并通过氧化应激水平(谷胱甘肽GSH、超氧歧化酶SOD及丙二醛MDA)、氧化还原酶的基因水平(谷胱甘肽巯基转移酶GST及SOD)及ABC转运蛋白(多药耐药蛋白MDR1及多药耐药相关蛋白MRP1)的变化,确定其中的主要机理。【结果】量子点引发的过氧化压力远大于同等浓度的游离Cd^2+;3种中药成分中,甘草酸具有良好的减毒作用,水飞蓟宾效果较弱,熊去氧胆酸则未见效果,该解毒作用主要依赖于过氧化应急压力的降低及抗氧化酶表达水平的上升;细胞转运蛋白的降低提示中草药可作为多药耐药肿瘤的有效治疗成分。【结论】中药成分可作为量子点生物医药应用中的重要配伍成分。[Objective]To study the interaction between three reducing traditional Chinese medicine ingredients and MPA-CdTe quantum dots in breast cancer(SK-BR-3)cells,and to unveil related mechanisms[Methods]Glycyrrhizic acid,silybin and ursodeoxycholic acid were employed as representative Chinese medicine ingredients to study quantum dots toxicity.Levels of oxidative stress(reduced glutathione,GSH;superoxide dismutase,SOD;malondialdehyde,MDA),gene expressions of anti-oxidative enzymes(Glutathione S-transferase,GST and SOD),and ABC transporters(multidrug resistance protein,MDR1 and multidrug resistance associated protein,MRP1)are to be detected in order to determine the related mechanism.[Results]The oxidative stress caused by quantum dots was much greater than that of free Cd^2+at the same concentration.The attenuation of the quantum dot toxicity for the three components depended on the reduction of the peroxidation emergency pressure.At the same time,the observed reduction of ABC transporters indicated that Chinese herbal medicine ingredients could be applied as potential inhibitors of multidrug resistance.[Conclusion]Traditional Chinese medicines can be developed as co-treatment drugs for quantum dots.
