摘 要:以云南、广东的广藿香为研究对象,采用气相色谱-质谱(GC-MS)联用技术对其挥发油的化学成分进行定性和定量分析,通过SIMCA+P进行主成分分析,利用RAPD技术进行分子学鉴定。GC-MS分析表明:从云南、广东的广藿香挥发油中分别鉴定出了48种、33种化学成分,其中30个成分相同,其主要成分基本一致,均为百秋李醇、苍术素、反式石竹烯。但其挥发油中百秋李醇含量差异较明显,云南广藿香中为28.53%,广东广藿香中为42.28%,相差13.75%。主成分分析表明:云南、广东的广藿香挥发油存在一定的差异。RAPD分析表明:虽然不同产地广藿香的品种亲缘关系较近,但筛选出了6条特异性引物,即OPB11、S227、OPD13、OPD70、OPD19、OPA07,可鉴别与区分云南、广东产地的广藿香,为广藿香提供了分子学鉴定依据。该研究为广藿香的可持续开发提供了应用基础。Using Pogostemon cablin from Yunnan and Guangdong as research objects,we qualitatively and quantitatively analyzed the chemical constituents of the volatile oil of Pogostemon cablin by gas chromatography-mass spectrometry(GC-MS).Moreover,we carried out the principal component analysis by SIMCA+P,and carried out the molecular identification by RAPD.GC-MS analysis results indicate that 48 and 33 chemical constituents are identified from the volatile oil of Pogostemon cablin from Yunnan and Guangdong,respectively,of which 30 are the same,and their main components are basically the same,all of which are patchouli alcohol,atractylodin,and trans-caryophyllene.However,there is a significant difference in the content of patchouli alcohol in the volatile oil,with 28.53% in Pogostemon cablin from Yunnan and 42.28% in Pogostemon cablin from Guangdong,with a difference of 13.75%.According to the principal component analysis,there are some differences in the volatile oil of Pogostemon cablin from Yunnan and Guangdong.PARD analysis shows that although there are close relationships among the varieties of Pogostemon cablin from different habitats,6 specific primers,including OPB11,S227,OPD13,OPD70,OPD19,and OPA07,can be screened out,which can identify and distinguish Pogostemon cablin from Yunnan and Guangdong,and provide a molecular identification basis for Pogostemon cablin.This study provides the application foundation for the sustainable development of Pogostemon cablin.
