摘 要:目的本研究模拟暴露于七氟烷废气的工作坏境,探讨长期吸入低浓度七氟烷对雌性小鼠早期胚胎及妊娠结果的影响。方法雌性昆明小鼠24只,雄性昆明小鼠12只,6周龄,体质量(20±2)g,分笼饲养。24只雌鼠随机分为A、B组,A组每天持续吸入0.1%七氟烷6 h,B组正常饲养。30 d后取发情期雌鼠与性成熟雄鼠2∶1合笼,合笼第2日晨7时观察雌鼠是否受孕。两组受孕小鼠在孕3.5 d取桑葚胚或囊胚,倒置显微镜下观察形态,孕18.5 d取胎鼠,测量体质量及身长。同时观察孕鼠有无流产现象。结果A组2只小鼠出现假孕现象,B组1只出现。孕3.5 d A组小鼠解剖发现子宫肿胀(宫腔未见胚胎)、异常胚胎现象、阴道流血、异常胎盘现象,B组未发现。小鼠孕18.5 d解剖得胚胎,A组胚胎体质量、身长均低于B组(P<0.05)。结论长期吸入0.1%七氟烷对小鼠早期胚胎发育有毒性作用,造成桑葚胚及囊胚发育异常,孕18.5 d胎鼠体质量及身长减小,同时引起孕鼠流产。Objective This study imitated the working environment of long-time exposure to low-concentration sevoflurane.To investigate the effect of low concentration sevoflurane on early embryonic development and pregnancy results of female mice.Method Twenty-four female KM mice and twelve male KM mice,aged 6 weeks,weighting(20±2)g,were fed separately.Twenty-four female rats were randomly divided into groups A and B.Group A continued to inhale 0.1%sevoflurane for 6 h every day,and group B was reared normally.Thirty days later,the estrous females and the sexually mature males were taken together at 2:1.The females were observed at 7 am on the second day to see if they were pregnant.Mulberry embryos or blastocysts were taken at 3.5 days of pregnancy from two groups of pregnant mice,and their morphology was observed under an inverted microscope.Fetal rats were taken at 18.5 days of pregnancy,and their body weight and length were measured.At the same time,the pregnant rats were observed for abortion.Results False pregnancy occurred in 2 mice in group A and 1 mouse in group B.At 3.5 days of pregnancy,the mice in group A were dissected and found uterine swelling(no embryos were seen in the uterine cavity),abnormal embryo phenomenon,vaginal bleeding,and abnormal placental phenomenon,but group B was not found.The embryos were dissected at 18.5 days of pregnancy.The body weight and length of the embryos in group A were lower than those in group B(P<0.05).Conclusion Long-term inhalation of 0.1%sevoflurane had toxic effects on early embryo development in mice,causing abnormal development of mulberry embryos and blastocysts.At 18.5 days of gestation,the body weight and length of fetal rats were reduced,and abortions occurred.
