摘　　要：斜纹夜蛾核多角体病毒(Spodoptera litura multicapsid nucleopolyhedrovirus,SpltMNPV)ORF75(Splt75)是杆状病毒核心基因。通过PCR方法、克隆、测序技术和生物信息学分析工具对Splt75基因及其编码蛋白Splt75进行了研究。结果表明,PCR扩增序列大小为363 bp,与Genbank登录的SpltMNPV G2株基因组全序列(AF325155)中的ORF75基因全长序列同源性100%;Splt75基因编码120个氨基酸,编码蛋白分子量为13137.16 Da,蛋白理论等电点为7.86,是稳定的疏水蛋白,存在跨膜结构区,属于ORF78(Ac78)超家族成员;Splt75蛋白中可能含有1个cAMP和cGMP依赖的蛋白激酶磷酸化位点、1个N-糖基化位点、1个酪蛋白激酶Ⅱ磷酸化位点、1个N-豆蔻酰化位点、1个蛋白激酶C磷酸化位点以及1个酰胺化位点;Splt75的二级结构以α螺旋和无规则卷曲为主要构成元件。Splt75基因及其编码蛋白Splt75的生物信息学分析结果为进一步研究其功能和作用机制提供了理论依据。Spodoptera litura multicapsid nucleopolyhedrovirus(SpltMNPV)ORF75(Splt75)is one of the baculovirus core genes.In order to investigate the possible role of Splt75 in the baculovirus life cycle,the ORF of Splt75 gene was cloned by PCR method from SpltMNPV genomic DNA,then sequenced using gene sequencing and analyzed by bioinformatics tools.The results showed that the ORF of Splt75 was 363 bp in length,encoding 120 amino acid residues,shared 100%identity with the sequence of Splt75 of SpltMNPV G2 in the GenBank(Genbank accession No.AF325155).The molecular weight of the predicted protein from Splt75 was 13137.16 Da,and the theoretical isoelectric point was about 7.86.Splt75 is a stable hydrophobic protein with transmember region.The prediction of the conserved domain indicated that Splt75 probably belonged to ORF78(Ac78)superfamily.Splt75 contained 1 ASN_GLYCOSYLATION,CAMP_PHOSPHO_SITE,MYRISTYL,PKC_PHOSPHO_SITE,CK2_PHOSPHO_SITE and AMIDATION.The secondary structure of Splt75 is mainly composed of alpha helix and random coil.These results provide theoretical basis for further study on the function and molecular mechanism of Splt75.
