摘  要:目的研究碳源、氮源和IAA等对金铁锁毛状根的侧根发生频率、空间排布和平均长度的影响,揭示毛状根空间构型和生长的关系。方法在悬浮培养条件下,通过改变培养基中碳源、氮源和IAA浓度,观察和分析金铁锁毛状根空间形态和生物量的变化。结果当培养基中添加30g/L蔗糖、NO3^-/NH4^+为28∶2(氮总浓度为60mmol/L)和0.5mg/L IAA时,单位长度侧根生长数、根的生长点数量、根的生长级数等参数均达到最佳,毛状根的长势较好,生物量最大。结论通过改变培养基中碳源、氮源和生长素浓度,获得有利于金铁锁毛状根生长的空间构型,促进毛状根生物量积累。Objective To investigate the effects of carbon source, nitrogen source and IAA on the lateral root formation, pattern and average length from the explant of hairy root, to dissect the relationship between pattern of lateral root development and growth of hairy root. Methods The changes in the architecture and biomass of suspension cultured hairy roots were observed and analyzed caused by modifications of carbon source, nitrogen source and IAA concentration in media.Results At the concentrations of 30 g/L carbon source, NO3^-/NH4^+ 28:2(total nitrogen source of 60 mmol/L), and 0.5 mg/L IAA in the media, all three parameters of number of lateral roots per unit length, number of root growth points, and the growth series of roots reached their peak point, especially this architecture of hairy roots system was beneficial for the accumulation of biomass. Conclusion At the optimized concentrations of carbon source, IAA,and NO3^-/NH4^+ ratio, the preferred architecture of hairy root system could be realized, by which accumulation of biomass was improved.
