摘 要:目的探讨枳术丸治疗慢传输型便秘脾虚证的可能作用机制。方法57只小鼠随机分成正常组15只,模型组、枳术丸组和莫沙必利组各14只,除正常组外其余各组小鼠采用番泻叶灌胃、低纤维饮食、限食和限水法制备慢传输型便秘脾虚证小鼠模型。造模成功后,枳术丸组按9.0g/(kg·d)灌胃,莫沙必利组按2.5mg/(kg·d)灌胃,模型组及正常组给予0.1ml/10g蒸馏水灌胃,每天2次,连续7天。观察各组小鼠一般行为学、结肠黏膜病理改变,检测小鼠首粒粪便排出时间、6h排便数量及质量,血清钙离子浓度及在体结肠肌电慢波的变化。结果正常组小鼠反应敏锐,被毛浓密、柔软,食纳佳,大便正常。与正常组比较,模型组小鼠反应迟钝,外形瘦小,食纳不佳,大便干硬、量少、颗粒细小,首粒粪便排出时间明显延长,6h排便数量及质量均明显减少,血清钙离子浓度明显降低,慢波频率明显增快、振幅明显减小(P<0.01);与模型组比较,枳术丸组和莫沙必利组小鼠反应较敏锐,被毛柔软,食纳佳,大便成形,首粒粪便排出时间明显缩短,6h排便数量及质量明显增加,血清钙离子浓度明显升高,慢波频率明显减慢、振幅明显增大(P<0.05或P<0.01)。与莫沙必利组比较,枳术丸组小鼠血清钙离子有所升高,慢波振幅增大(P<0.05)。结论枳术丸能改善慢传输型便秘脾虚证小鼠排便情况,可能通过提高血清钙离子的浓度,从而调节其结肠慢波,增强肠道动力。Objective To explore the possible mechanism of Zhizhu Pills(枳术丸)in the treatment of slow transit constipation(STC)with spleen deficiency syndrome.Methods A total of 57 mice were randomly divided into a normal groups with 15 mice,and a the model group,a Zhizhu Pills group and a mosapride group with 14 mice in each group.Mice except for the normal group were given senna leaf for gavage,low-fiber diet,diet restriction and water restriction to prepare mice model STC and spleen deficiency syndrome.After successful modeling,the Zhizhu Pills group was given 9.0 g/(kg·d)by gavage,the mosapride group was given 2.5 mg/(kg·d)by gavage,the model group and the normal group was given 0.1 ml/10 g distilled water by gavage,2 times a day for 7 consecutive days.We observed the general behavior and pathological changes of colonic mucosa of each group of mice,and detected the time of the first feces excretion,the quantity and quality of 6 h defecation,the serum calcium ion concentration and the changes of colonic myoelectric slow wave in vivo.Results The mice in the normal group reacted sensitively,with thick and soft fur on the back,good food appetite,and normal stools.Compared with the normal group,the mice in the model group were sluggish,skinny,poor in appetite,dry,hard and less stools with small particles.The first stool discharge time was significantly prolonged,and the number and quality of stools in 6 hours were significantly reduced.Serum calcium ions concentration was significantly reduced,the slow wave frequency was significantly increased,and the amplitude was significantly reduced(P<0.01).Compared with the model group,the mice in the Zhizhu Pills group and the mosapride group had sensitive responses,with soft fur on the back,good appetite and normal stool.The first fecal discharge time was significantly shortened,the number and quality of the 6 h defecation were significantly increased,the serum calcium ion concentration was significantly increased,the slow wave frequency was significantly slowed down,and the am
