摘 要:目的研究苦参碱对神经病理性疼痛大鼠P2X2和P2X3受体表达的影响。方法将24只SD大鼠随机分为对照组、模型组和实验组,每组各8只。将模型组和实验组大鼠用坐骨神经慢性压迫损伤法制备神经病理性疼痛大鼠模型;假手术组仅游离坐骨神经但不结扎。建模后,实验组按30 mg·kg^-1·d^-1的剂量腹腔注射苦参碱溶液,qd;假手术组和模型组均给予腹腔注射等量0.9%NaCl,qd。3组大鼠连续干预14 d。比较3组大鼠的热缩足反射潜伏期(TWL)和机械缩足反射阈值(MWT),并用荧光定量聚合酶链反应测定背根神经节P2X2和P2X3 mRNA的表达情况。结果干预14 d后,假手术组、模型组和实验组大鼠TWL分别为(19.25±2.12),(6.75±2.12)和(17.25±2.25)s,MWT分别为(38.13±1.81),(14.03±2.14)和(36.25±2.60)g,背根神经节P2X2 mRNA表达量分别为1.04±0.20,2.28±0.65和1.23±0.27,P2X3 mRNA表达量分别为0.95±0.17,1.64±0.52和1.09±0.21,模型组的上述指标与假手术组和实验组比较,差异均有统计学意义(均P<0.05)。结论苦参碱可通过抑制神经病理性疼痛大鼠P2X2和P2X3受体的表达,从而减轻痛觉过敏。Objective To study the effect of matrine on the expressions of P2X2 and P2X3 receptors in rats with neuropathic pain.Methods A total of 24 Sprague-Dawley rats were randomly divided into sham-operation,model and experimental groups with 8 rats per group.Rats in model and experimental groups were built neuropathologic pain rat model by chronic compression injury of sciatic nerve,while rats in sham-operation group,the sciatic nerve was free but not ligated.The experimental group was intraperitoneally injected with matrine solution at the dose of 30 mg·kg^-1·d^-1,qd.Sham-operation and model groups were intraperitoneally injected the equal volume of 0.9%NaCl,qd.Three groups were treated for 14 days.The thermal withdrawal latency(TWL)and mechanical withdrawal threshold(MWT)were compared among three groups,and the mRNA expressions of P2X2 and P2X3 were measured by reverse transcription-polymerase chain reaction.Results 14 days after intervention,the main indexes in sham-operation,model and experimental groups were compared:TWL values were(19.25±2.12),(6.75±2.12)and(17.25±2.25)s,MWT values were(38.13±1.81),(14.03±2.14)and(36.25±2.60)g,P2X2 mRNA in dorsal root ganglion were 1.04±0.20,2.28±0.65 and 1.23±0.27,P2 X3 mRNA in dorsal root ganglion were0.95±0.17,1.64±0.52 and 1.09±0.21.The above-mentioned indexes in the model group were significantly different from those in the sham-operation group and experimental group(all P<0.05).Conclusion Matrine can relieve hyperalgesia by inhibiting the expressions of P2X2 and P2X3 receptors in rats with neuropathic pain.
