摘 要:目的探讨百香果籽油对帕金森病(PD)小鼠星型胶质细胞活化的影响。方法用腹腔注射30 mg·kg^-11-甲基-4-苯基-1,2,3,6-四氢吡啶建立PD小鼠模型,并随机分为模型组和低、高剂量实验组,每组12只;另取12只正常小鼠作为对照组。低、高剂量实验组分别灌胃给予50和100 mg·kg^-1百香果籽油,qd;模型组和对照组均灌胃给予等体积的蒸馏水,qd。4组小鼠均连续给药42 d。用转棒实验和悬挂实验检测小鼠的行为学能力,用免疫组织化学法检测海马和额叶皮质胶质纤维酸性蛋白(GFAP)表达,用分光光度计检测血清炎性因子的含量。结果对照组、模型组和低、高剂量实验组的滚动圈数分别为(69.25±2.30),(38.75±2.16),(52.38±2.23)和(57.21±2.20)圈,悬挂时间分别为(40.53±3.28),(17.36±1.42),(26.45±2.35)和(31.34±2.78)s,肿瘤坏死因子-α分别为(24.27±3.35),(58.62±4.93),(39.14±3.48)和(32.25±5.03)nmol·L^-1,白细胞介素-1β分别为(9.25±1.64),(29.36±2.61),(20.54±1.93)和(13.73±1.18)nmol·L^-1,白细胞介素-6分别为(11.57±2.85),(38.48±5.03),(22.62±3.19)和(12.92±3.62)nmol·L^-1,海马GFAP阳性细胞数目分别为(21.44±3.19),(78.83±5.99),(57.83±4.74)和(41.78±3.10)个,额叶皮质GFAP阳性细胞数目分别为(35.83±3.19),(97.72±7.84),(69.33±5.20)和(60.39±5.49)个,模型组的上述指标与对照组和低、高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论百香果籽油可通过抑制星型胶质细胞活化,从而改善PD小鼠的行为。Objective To explore the effect of passion fruit seed oil on the behavior and activation of astrocytes in mice with Parkinson’s disease(PD).Methods Mice were intraperitoneally injected with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)to construct PD model.After the model was successfully prepared,the mice were randomly divided into model group,experimental-L group and experimental-H group with 12 cases per group.Another 12 normal mice were taken as the control group.The experimental-L and experimental-H groups were given 50 and 100 mg·kg^-1 passion fruit seed oil with intragastric administration,qd,respectively.The model and control groups were given the same volume of distilled water with intragastric administration,qd.Four groups were treated for 42 d.Rotation experiment and suspension experiment were used to detect the behavior ability of mice.Spectrophotometer was utilized to examine the content of inflammatory factors in serum.Immunohistochemical test was given to check the expression of glial fibrillary acidic protein(GFAP)in hippocampus and frontal cortex.Results The rotarod cycles of control group,model group,experimental-L group and experimental-H group were(69.25±2.30),(38.75±2.16),(52.38±2.23)and(57.21±2.20)cycles,the suspension time were(40.53±3.28),(17.36±1.42),(26.45±2.35)and(31.34±2.78)s,the tumor necrosis factor-αwere(24.27±3.35),(58.62±4.93),(39.14±3.48)and(32.25±5.03)nmol·L^-1,the interleukin-1 beta were(9.25±1.64),(29.36±2.61),(20.54±1.93)and(13.73±1.18)nmol·L^-1,the interleukin-6 were(11.57±2.85),(38.48±5.03),(22.62±3.19)and(12.92±3.62)nmol·L^-1,the number of GFAP-positive cells in hippocampus were(21.44±3.19),(78.83±5.99),(57.83±4.74)and(41.78±3.10),the number of GFAP-positive cells in frontal cortex were(35.83±3.19),(97.72±7.84),(69.33±5.20)and(60.39±5.49).Comparing with the model group,the above indicators in the other groups were all with significant difference(all P<0.05).Conclusion Passion fruit seed oil could improve the behavior of mice with PD
