摘 要:目的观察电针手厥阴心包经穴对脑缺血后大鼠神经生长抑制因子及其受体表达的影响,探讨电针心包经穴对大鼠脑缺血后神经修复的作用机制。方法将SD大鼠随机分为正常组、假手术组、模型组、心包经组和肺经组,每组18只。颈外动脉插入线栓法制作局灶性脑缺血模型。成模后第2日起电针干预30 min,每日1次,干预6 d休息1 d,分别于7、14、21 d进行取材,每个时间点6只。ELISA检测血清神经生长抑制因子(Nogo-A)含量,RT-qPCR检测脑梗死区Nogo-A及其受体NgR1的mRNA表达。结果与同一时间点正常组和假手术组比较,模型组大鼠血清Nogo-A含量及脑梗死区Nogo-A和NgR1 mRNA表达各时间点均明显升高(P<0.01);与同一时间点模型组比较,心包经组大鼠血清Nogo-A含量第21日及脑梗死区Nogo-A和NgR1 mRNA表达第7、21日明显降低(P<0.01),肺经组大鼠脑梗死区NgR1 mRNA表达第7日明显降低(P<0.05),NgR1 mRNA表达第14日明显升高(P<0.01);与同一时间点肺经组比较,心包经组大鼠血清Nogo-A含量第21日明显降低(P<0.05),脑梗死区Nogo-A和NgR1 mRNA表达各时间点明显降低(P<0.01)。结论电针心包经穴可促进脑缺血后神经修复,其机制与下调模型大鼠血清Nogo-A含量、抑制缺血侧脑组织Nogo-A和NgR1的mRNA表达相关。Objective To observe the effects of electroacupuncture(EA)stimulation of acupoints of pericardium meridian on Nogo-A and NgR1 expression in cerebral ischemia rats;To explore the mechanism of EA at the pericardium meridian on nerve repair after cerebral ischemia in rats.Methods The SD rats were randomly divided into normal control group,sham-operation group,model group,pericardium meridian,and lung meridian group,with 18 rats in each group.The model of focal cerebral ischemia was selected and made by external carotid artery insertion thread embolization method.From the second day after successful modeling,EA intervention was performed for 30 minutes,once a day,intervention for 6 days and rest for 1 day.Serum and brain tissue were obtained on 7 th,14 th and 21 st day,with 6 rats at each time point.Serum nerve growth inhibitory factor(Nogo-A)was detected by ELISA.RT-qPCR was used to detect the expression of Nogo-A and its receptor NgR1 mRNA in cerebral infarction area.Results Compared with the normal control group and sham-operation group,the serum Nogo-A content and the expression of Nogo-A and NgR1 mRNA in the cerebral infarction area in the model group significantly increased at each time point(P<0.01).Compared with the model group at the same time point,the serum Nogo-A content and the expressions of Nogo-A and NgR1 mRNA in the cerebral infarction area on the 7 th and 21 st days in the pericardium meridian group were significantly reduced on the 21 st day(P<0.01),the expression of NgR1 mRNA in the cerebral infarction area of the lung meridian group was significantly reduced on the 7 th day(P<0.05),and NgR1 mRNA expression increased significantly on the 14 th day(P<0.01).Compared with the lung meridian group at the same time point,the serum Nogo-A content in the pericardium meridian group was significantly reduced on the 21 st day(P<0.05),and the Nogo-A and NgR1 mRNA expression in the cerebral infarction area was significantly reduced at each time point(P<0.01).Conclusion EA stimulation of acupoints of pericardiu
