摘 要:通过观察盐地碱蓬对2型糖尿病(T2DM)大鼠血糖及胰腺Bax、Bcl-2、Caspase-3蛋白表达的影响,探讨盐地碱蓬改善糖代谢的作用机制。采用高脂饲料联合腹腔注射链脲佐菌素(STZ)建立2型糖尿病大鼠模型,检测空腹血糖(FBG)、餐后2 h血糖(PG2h)、糖耐量(OGTT)并计算曲线下面积(AUC);检测血清中糖化血红蛋白(GHb)、C-肽(C-P)并计算胰岛素抵抗指数(IR指数);苏木素-伊红(HE)染色观察胰腺组织病理形态学变化;免疫组化检测(IHC)胰腺组织中Bax、Bcl-2蛋白平均光密度值,蛋白免疫印迹法(Western blot)检测胰腺组织中Bax、Bcl-2、Caspase-3、cleaved-Caspase-3蛋白表达水平。结果发现与正常组相比模型组FBG、PG2h、OGTT-AUC及GHb显著升高,C-P显著降低(P<0.01),胰腺组织中Bcl-2蛋白表达水平显著下调(P<0.01),Bax、Caspase-3、cleaved-Caspase-3蛋白水平显著上调(P<0.01);与模型组相比,盐地碱蓬组FBG、PG2h、OGTT-AUC及GHb显著降低,C-P显著升高(P<0.01),胰腺组织中Bcl-2蛋白表达水平显著上调,Bax、Caspase-3、cleaved-Caspase-3蛋白表达水平显著下调(P<0.01)。研究表明盐地碱蓬可以改善T2DM大鼠糖代谢,其作用机制可能与上调胰腺组织中Bcl-2,下调Bax、Caspase-3,降低cleaved-Caspase-3蛋白表达水平有关。To observe the effects of Suaeda salsa on the blood glucose and expressions of Bax,Bcl-2 and Caspase-3 in the pancreas of type 2 diabetic(T2DM)rats,and to explore the mechanism of improving the metabolism of sugar by S.salsa.A type 2 diabetic rat model was built by combining high-fat diet with streptozotocin(STZ),the fasting blood glucose(FBG),plasma glucose-2 h(PG2h),oral glucose tolerance test(OGTT)and area under the curve(AUC)were measured.The glycosylated hemoglobin(GHb)and C-peptide(C-P)in the serum were tested and insulin resistance index(IR index)was calculated.Pathomorphological changes in the pancreatic tissues were observed by means of hematoxylin and eosin(HE)staining.The mean optical density of Bax and Bcl-2 proteins in the pancreatic tissues were tested by immunohistochemistry(IHC),and the expression levels of Bax,Bcl-2,Caspase-3 and cleaved-Caspase-3 proteins in the pancreatic tissues were tested by Western blot.The results showed that compared with the normal group,the FBG,PG2h,OGTT-AUC and GHb of the model group were significantly increased,C-P was significantly decreased(P<0.01)and the expression of Bcl-2 protein in the pancreatic tissues was significantly down-regulated(P<0.01),while the expressions of Bax,Caspase-3 and cleaved-Caspase-3 proteins were significantly up-regulated(P<0.01).Compared with the model group,FBG,PG2h,OGTT-AUC and GHb were significantly decreased,while C-P was significantly increased in the Suaeda salsa group.The expression of Bcl-2 protein in the pancreatic tissues was significantly up-regulated,while the expressions of Bax,Caspase-3 and cleaved-Caspase-3 proteins were significantly down-regulated(P<0.01).The present study showed that Suaeda salsa can improve the glucose metabolism in T2DM rats,and its mechanism may be related to the up-regulation of Bcl-2,the down-regulation of Bax and Caspase-3,and the decrease of the expression of cleaved-Caspase-3 protein in the pancreatic tissues.
