摘 要:目的探究丹参酮ⅡA作用于成纤维细胞预防SD大鼠椎板切除术后硬膜外疤痕增生的机制。方法选取L1椎板切除SD大鼠24只,按照随机数字表分为Ⅰ~Ⅳ组,每组各6只。Ⅰ组腹腔注射100 mg/kg丹参酮ⅡA,Ⅱ组腹腔注射200 mg/kg丹参酮ⅡA,Ⅲ组腹腔注射300 mg/kg丹参酮ⅡA,Ⅳ组腹腔注射等体积生理盐水。观察术后各组大鼠伤口愈合情况,通过影像学方法计算瘢痕面积,利用流式细胞仪检测成纤维细胞凋亡率和细胞周期G0/G1期比例,通过Western blot技术检测各组大鼠疤痕组织中p53、survivin、Bcl-2、Bax和Caspase-3水平。结果术后所有大鼠伤口愈合良好,干预后1个月,4组瘢痕面积差异有统计学意义(P<0.05),由大到小依次为Ⅳ组、Ⅰ组、Ⅱ组和Ⅲ组。4组干预后24 h、72 h及120 h的成纤维细胞凋亡率和G0/G1期比例差异及3个时间点的组间差异均有统计学意义(P<0.05)。干预后120 h,4组瘢痕组织中p53、survivin、bcl-2、bax和Caspase-3水平差异均有统计学意义(P<0.05)。结论丹参酮ⅡA具有抑制成纤维细胞增殖、诱导凋亡的作用,其效果存在浓度依赖性,机制可能与多种细胞因子的调节有关。Objective To explore the effects of tanshinoneⅡA on fibroblasts in preventing epidural scar proliferation after laminectomy in SD rats.Methods Twenty-four SD rats with L1 laminectomy were randomly divided into 4 groups,with 6 rats in each group.GroupⅠreceived intraperitoneal injection of 100 mg/kg tanshinoneⅡA,groupⅡreceiving 200 mg/kg tanshinoneⅡA,groupⅢreceiving 300 mg/kg tanshinoneⅡA,and groupⅣreceiving equal volume of saline.The wound healing of the rats in each group was observed.Scar area was calculated by imaging method.The apoptosis rate of fibroblast and cell cycle G0/G1 phase ratio were detected by flow cytometry.The contents of p53,survivin,Bcl-2,Bax and Caspase-3 in the scar tissue of the rats in each group were detected by Western blot.Results All the wounds healed well after operation.There was significant difference in scar area between the four groups one month after intervention(P<0.05).The scar area was biggest in groupⅣ,followed by groupⅠ,groupⅡand groupⅢ.The apoptotic rate of fibroblasts and the ratio of G0/G1 phase were significantly different between different time points in the four groups(P<0.05).The apoptotic rate of fibroblasts and the ratio of G0/G1 phase were different among the four groups(P<0.05).The expression of p53,survivin,Bcl-2,Bax and Caspase-3 in the scar tissue 120 h after intervention showed statistical differences among the four groups(P<0.05).Conclusions TanshinoneⅡA can inhibit the proliferation of fibroblasts and induce apoptosis.The effect is concentration-dependent.The mechanism may be related to the regulation of various cytokines.
