摘 要:目的:探讨刺芒柄花素(FNN)对单侧输尿管梗阻(UUO)大鼠肾脏间质纤维化的影响。方法:80只SD大鼠随机分为假手术(sham)组、模型(model)组、低剂量FNN干预(L-FNN)组和高剂量FNN干预(H-FNN)组,每组20只。HE染色观察肾组织病理变化,Masson染色观察肾间质纤维化程度。Western blot检测蛋白表达。结果:Sham、model、L-FNN和H-FNN组肾小管损伤评分分别为0.08±0.02、4.22±0.74、3.06±0.52和2.24±0.31,四组间差异有统计学意义(P<0.01)。Sham、model、L-FNN和H-FNN组肾间质纤维化百分比分别为(13.14±1.02)%、(82.15±12.36)%、(57.28±11.23)%和(26.15±3.76)%,四组间差异有统计学意义(P<0.01)。L-FNN和H-FNN组TGF-β1和Smad3表达显著低于model组(P<0.01),H-FNN组TGF-β1和Smad3表达又显著低于L-FNN组(P<0.01)。结论:刺芒柄花素抑制TGF-β1/Smad3通路,可以抑制UUO大鼠肾脏间质纤维化的进程。Objective:To explore therapeutic effect of formononetin(FNN)on renal interstitial fibrosis in unilateral ureteral obstmction(UUO)rats.Methods:80 rats were devided into sham group(n=20),model group(n=20),low dose FNN(L-FNN,n=20)group and high dose FNN(H-FNN,n=20)group.HE and Masson staining was used to observe renal pathological change.The protein was detected by Western Blot analysis.Results:The score of renal tubular injury score in sham,model,L-FNN,and H-FNN group was 0.08±0.02,4.22±0.74,3.06±0.52,and 2.24±0.31.There was significant difference among the four groups(P<0.01).The percent of renal interstitial fibrosis in sham,model,L-FNN,and H-FNN group was(13.14±1.02)%,(82.15±12.36)%,(57.28±11.23)%,and(26.15±3.76)%.There was significant difference among the four groups(P<0.01).The protein expression of TGF-β1 and Smad3 in L-FNN,and H-FNN group was significantly lower than that in model group(P<0.01),and which in H-FNN group was significantly lower than that in L-FNN group(P<0.01).Conclusion:Formononetin can inhibit TGF-β1/Smad3 signaling pathway to antagonize renal interstitial fibrosis in UUO rats.
