摘  要:目的建立龙胆和坚龙胆药材的HPLC指纹图谱,并结合化学计量学分析法对龙胆和坚龙胆进行鉴别及质量评价。方法采用HPLC对不同产地的20批龙胆和17批坚龙胆进行指纹图谱研究,建立龙胆和坚龙胆的对照指纹图谱,采用液质联用技术对龙胆和坚龙胆指纹图谱共有峰进行指认,并结合聚类分析和主成分分析对龙胆和坚龙胆进行鉴别及质量评价。结果龙胆指纹图谱有13个共有峰,坚龙胆指纹图谱有10个共有峰,其中3个峰分别为马钱苷酸、獐牙菜苦苷和龙胆苦苷;龙胆和坚龙胆的指纹图谱存在明显差异,利用坚龙胆的共有峰7能够对二者进行鉴别;根据共有峰的聚类分析和主成分分析结果,将龙胆和坚龙胆分为3类;从主成分得分来看,龙胆药材质量优于坚龙胆。结论本研究建立的方法能高效、准确鉴别龙胆和坚龙胆药材,并有效对其质量进行评价。Objective To establish HPLC fingerprints of Gentiana scabra Bge.and Gentiana rigescens Franch;To identify and conduct quality evaluation for Gentiana scabra Bge.and Gentiana rigescens Franch combined with chemometrics.Methods HPLC method was used to study the fingerprints of 20 batches of Gentiana scabra Bge.and 17 batches of Gentiana rigescens Franch.from different producing areas.The control fingerprints of Gentiana scabra Bge.and Gentiana rigescens Franch.were established.The common peaks were identified by LC-MS.The hierarchical clustering analysis and principal components analysis were used to identify and evaluate the quality of Gentiana scabra Bge.and Gentiana rigescens Franch.Results There were 13 common peaks in the fingerprint of Gentiana scabra Bge.and 10 common peaks in the fingerprint of Gentiana rigescens Franch.Three of these peaks were identified as loganic acid,swertiamarin and gentiopicroside respectively.The fingerprints of Gentiana scabra Bge.and Gentiana rigescens Franch.were obviously different.The common peak 7 of Gentiana rigescens Franch.could be used to identify and distinguish Gentiana scabra Bge.and Gentiana rigescens Franch.According to the results of cluster analysis and principal component analysis,Gentiana scabra Bge.and Gentiana rigescens Franch.were divided into three major groups,and the quality of Gentiana scabra Bge.was better than that of Gentiana rigescens Franch.Conclusion This method can identify and distinguish Gentiana scabra Bge.and Gentiana rigescens Franch.quickly and accurately,which can successfully evaluate the quality of Gentiana scabra Bge.and Gentiana rigescens Franch.
