藏紫草的本草考证及质量研究  

Herbal authentication and quality study on Radix Onosmatis

摘　　要：对采自西藏不同地区的59批藏紫草样品的高效液相(HPLC)指纹图谱进行相似度比较、聚类分析和主成分分析,结合本草考证和野外调查,发现藏紫草的两种正品来源为团花滇紫草及细花滇紫草(长花滇紫草)。两种基源的种内差异较小,种间差异明显。团花滇紫草种内相似度在0.90~0.96之间,细花滇紫草及其变种长花滇紫草种内相似度在0.97~0.99之间;两个种之间的相似度不到0.8。聚类分析显示,团花滇紫草及细花滇紫草(长花滇紫草)之间距离较远;测定两者中β,β′-二甲基丙烯酰紫草素的含量,发现54批细花滇紫草中β,β′-二甲基丙烯酰紫草素的含量均低于定量限,几乎无法检测出来;而5批团花滇紫草中的含量在0.05%~0.20%之间,远高于细花滇紫草。因此,文章认为《晶珠本草》中所说的上品藏紫草为团花滇紫草,高效液相指纹图谱可以作为两种正品藏紫草基源的鉴别依据。It was found that the two authentic sources of Radix Onosmatis were Onoasma hookeri and Onoasma glomeratum by similarity comparison,cluster analysis and principal component analysis,combined with Herbal Authentication and field investigation with the high performance liquid chromatography(HPLC)fingerprints of59 batches of Radix Onosmatis samples from different regions of Tibet.The differences between the two species were obvious while the intraspecific differences were small.The intraspecific similarity of O.hookeri was between 0.90 and 0.96,while the intraspecific similarity of O.glomeratum was between 0.97 and 0.99,The similarity between the two species was less than 0.8.Cluster analysis also showed that the distance between O.hookeri and O.glomeratum was far.The contents ofβ’-β’--dim ethyl propene shikonin in 54 batches of O.hookeri were found to be lower than the quantification limit,and almost impossible to be detected,while the content of 5 batches of O.glomeratum was between 0.05%and 0.20%,which was much higher than those of O.hookeri.Therefore,it is believed that O.glomeratum was the top-quality herb mentioned in Jingzhu Materia Medica.HPLC fingerprint could be used as the basis for identification of the two authentic Radix Onosmatis.
