摘 要:目的:建立同时测定金银花中绿原酸、新绿原酸和异绿原酸含量的方法。方法:采用高效液相色谱法。色谱柱为Phenomenex Luna C18柱(5μm,4.6 mm×250 mm),流动相为0.1%甲酸水溶液-乙腈(梯度洗脱),检测波长240 nm,柱温25℃。结果:绿原酸、新绿原酸和异绿原酸在0.013~0.526 mg/mL内呈良好线性关系(r为0.9993);平均加样回收率分别为99.53%、99.73%、100.23%,RSD分别为4.27%、2.49%、1.58%(n=6)。结论:该方法可靠、稳定、重现性好,可作为金银花中绿原酸、新绿原酸和异绿原酸的含量测定方法。Objective:To establish a method for simultaneous determination of chlorogenic acid,neochlorogenic acid and isochlorogenic acid in honeysuckle.Methods:HPLC method was adopted.The determination was performed on phenomenex luna C18(5μm,4.6 mm×250 mm)column with mobile phase consisted of 0.1%formic acid solution-acetonitrile(gradient elution).The detection wavelength was set at 250 nm and the column temperature was set at 25℃.Results:The linear ranges of chlorogenic acid,neochlorogenic acid and isochlorogenic acid were all 0.013-0.526 mg/mL(r=0.9993).The average recoveries of three compounds were 99.53%(RSD=4.27%,n=6),99.73%(RSD=2.49%,n=6),and 100.23%(RSD=1.58%,n=6),respectively.Conclusion:The method is feasible,stable,good reproducibility,for the determination of chlorogenic aeid,neochlorogenic acid and isochlorogenic acid in honeysuckle.
