摘 要:目的探讨Roseburia hominis对肝硬化大鼠肝功能、血清细胞因子及肝脏、肠道组织的影响。方法30只体质量250~300 g的雄性大鼠,随机分为3组:RH组10只、正常对照组10只,模型对照组10只,正常对照组给予生理盐水腹腔注射,其余20只大鼠均用四氯化碳制备肝硬化大鼠模型。模型制备后,在处死大鼠之前15天开始,RH组大鼠给予R.hominis活菌菌液灌胃,模型对照组给予等量生理盐水灌胃,采集样本测定。观察肝功能及血清细胞因子;对肝脏和肠道组织行石蜡包埋、切片、苏木精和伊红染色,病理切片观察对比组织的炎症和损伤程度。结果RH组大鼠肝功能显著好转,ALT、AST、TB与模型组对比均显著下降(t=2.6331、2.7012、2.5814,P=0.0115、0.0089、0.0133)。血清细胞因子的检测结果显示,与模型对照组相比,RH组大鼠IFN-γ和IL-10的水平均显著升高(t=2.2562、2.0012,P=0.0258、0.0416)。RH组大鼠肝脏、肠道组织病理结果较模型对照组明显好转。结论R.hominis对肝硬化大鼠肝功能有改善作用,可减轻肝脏及肠道组织损伤,提高血清中IFN-γ和IL-10水平。Objective To explore the effects of Roseburia hominis on liver function,serum cytokines and the issue of liver and intestine in cirrhotic rats.Methods A total of 30 male SD rats(weighing 250-300 g)were randomly divided into RH group(n=10),normal control group(n=10),or model control group(n=10).Normal control group was given intraperitoneal injection of normal saline;the other two groups were given carbon tetrachloride to establish models of liver cirrhosis.15 days before the rats were put to death,the RH group was intragastrically administered viable R.hominis liquid,while the model control group was given equivalent amount of saline,then specimens were collected for determination of the contents of liver function and serum cytokines.The liver and intestinal tissues were taken for paraffin embedding,sectioning,and hematoxylin and eosin staining.Pathological sections were used to observe and compare the degrees of inflammation and injury between groups.Results The liver function of rats in RH group were significantly improved,and ALT,AST and TB significantly decreased compared with model controls(t=2.6331,2.7012,2.5814;P=0.0115,0.0089,0.0133).The levels of serum IFN-γand IL-10 in the RH group were significantly higher than in the model control group(t=2.2562,2.0012;P=0.0258,0.0416).The pathological results of liver and intestinal tissue in the RH group were significantly better than in the model control group.Conclusion R.hominis can improve liver function,reduce the damage of liver and intestinal tissue and improve the serum levels of IFN-γand IL-10 in cirrhotic rats.
