摘 要:本研究用线粒体细胞色素b(Mitochondrial cytochrome b,Cyt b)基因全序列分析了松嫩平原区黑龙江省连环湖(LHH)72尾和龙虎泡(LHP)49尾、吉林省查干湖(CGH)54尾、新立城水库(XLC)22尾和二龙山水库(ELS)29尾,共226尾大银鱼Protosalanx hyalocranius的群体遗传多样性。结果在1141 bp的Cyt b基因序列中检测到60个变异位点,其中单变异位点32个,简约信息位点28个;检测到38个单倍型,每个群体的单倍型数(nh)为5~16,LHH群体单倍型最多,而LHP群体最少。群体单倍型多样性(Hd)为(0.656±0.051)^(0.939±0.028),核苷酸多样性(Pi)为(0.00121±0.00010)^(0.00496±0.00150),XLC群体遗传多样性最高,LHP群体最低。分子方差分析(AMOVA)结果表明:大银鱼遗传差异主要来自群体内(93.81%),少数来自群体间(6.19%),除LHP和CGH群体间分化不显著外,其他群体间遗传分化均达到显著水平(P<0.05)。群体间遗传距离为0.0014~0.0040,LHP和LHH之间遗传距离最小(0.0014),吉林省境内的XLC和ELS群体间遗传距离最大(0.0040),CGH先与LHP和LHH聚为一类,再与ELS和XLC聚类。本研究结果可为松嫩平原区大银鱼的繁育和增殖等渔业管理提供遗传数据支持。In this study,the genetic diversity of clearhead icefish(Protosalanx hyalocranius)was investigated in 226 samples of five populations collected from Lianhuan Lake(LHH)(n=72)and Longhupao Lake(LHP)(n=49),Heilongjiang Province,and Chagan Lake(CGH)(n=54),Xinlicheng Reservoir(XLC)(n=22)and Erlongshan Reservoir(ELS)(n=29),Jilin Province at Songnen Plain by using the complete sequence of mitochondrial cytochrome b(Cyt b)gene.A total of 60 mutation sites were detected in the 1141 bp Cyt b gene sequence,including 32 singleton variable sites and 28 parsimony informative sites.38 haplotypes were detected,with the haplotype number(nh)ranging from 5 to 16 each population,with the maximal haplotype number in the LHH population,and the minimal haplotype number in LHP population.The haplotype diversity(Hd)was shown to be ranged from(0.656±0.051)to(0.939±0.028),and the nucleotide diversity(Pi)to be varying from(0.00121±0.00010)to(0.00496±0.00150).The maximal genetic diversity was observed in XLC population,and the minimal in LHP population.The molecular variance analysis(AMOVA)showed that the genetic differences in the clearhead icefish were primarily derived from intra populations(93.81%)and minority from inter populations(6.19%).Except for the difference between LHP and CGH populations,there was significantly genetic differentiation among other populations(P<0.05).The genetic distance between populations was ranged from 0.0014 to 0.0040,with the minimal genetic distance(0.0014)between LHP and LHH,and the maximal one(0.0040)between XLC and ELS although they were in the same province.It was found that CGH was clustered with LHP and LHH first,and then with ELS and XLC.The findings provide genetic data for the breeding and reproductive fishery management of clearhead icefish in Songnen Plain.
