摘 要:以基于酶切的简化基因组测序技术(GBS),测定了福建省南平市、三明市和龙岩市地区野生棘胸蛙Rana spinosa子一代3龄蛙群体66份样本的全基因组DNA,依据SNP的基因分型结果,采用邻近法构建系统演化树和主成分分析,以探究福建内陆山区棘胸蛙群体的遗传多样性。共获得66.79 Gb的测序数据,平均每个样本测序数据为1.01 Gb,经过筛选条件过滤,最后共获得8615个高质量的SNP位点,其中以龙岩群体的SNP最多,为3589个,南平群体的SNP最少,为2153个,说明不同地区棘胸蛙群体SNP数存在较大差异。利用系统发生树分析结果表明,在3个棘胸蛙群体中,南平群体能聚为一类,三明和龙岩群体未能明确分类。主成分分析结果表明,3个群体的区分度不明显,虽然在聚类时呈现一定程度的分化,但未形成一定的地理隔离。福建棘胸蛙群体之间存在大量的SNP。该研究可为棘胸蛙的识别和进化提供依据,为更好地保护与利用棘胸蛙种质资源提供参考。The whole genome DNA was analyzed in 66 samples of 3-year-old spine frog Rana spinosa progeny from frog population in Nanping,Sanming and Longyan areas of Fujian Province by reduced-representation genome sequencing(GBS)based on enzymatic digestion,and the genetic diversity of the spine frogs in the inland mountainous areas of Fujian province was established by phylogenetic tree built by using Neighbor-joining method and principal component analysis(PCA)by the proximity method based on the genotyping results of SNP.The results showed that a total of 66.79 Gb of sequences was generated,with average 1.01 Gb each sample.After screening,8,615 high quality SNP sites were retained,the most SNPs of 3,589 in Longyan group,and the least SNP of 2,153 in Nanping group,indicating that there were large differences in the number of SNPs in the spine frog populations in different regions.The phylogenetic tress revealed that the Nanping group was grouped together,and the Sanming and Longyan groups were not clearly classified.Principal component analysis showed that the discrimination of the three groups was not very obvious due to without formation of a certain geographical isolation,though they showed a certain degree of differentiation in clustering.There were a large number of SNPs between the spine frog populations.This finding can provide a basis for the identification and evolution of spiny frogs,and a reference for better protection and utilization of the frog germplasm resources in the future.
