摘  要:在水温(15±1)℃下,将体质量(4.1±0.6)g的花鳅Cobitis taenia随机分为6组,每组3个重复,每个重复30尾鱼,放入容水180 L的75 cm×60 cm×50 cm的聚乙烯塑料水族箱中,每箱CuSO4浓度为0 mg/L(对照组)、0.10 mg/L、0.17 mg/L、0.30 mg/L、0.51 mg/L和0.90 mg/L,采用静水生物测试法研究铜离子对花鳅的急性毒性,评价铜离子对花鳅的安全性。试验24 h时从各处理组中随机取3尾花鳅,测定鳃、肝中铜离子含量及肝组织过氧化物酶(POD)、谷氨酸脱氢酶(GDH)活性和脑组织中谷氨酸(Glu)浓度。结果表明,花鳅随铜离子浓度升高毒性逐渐增强,24 h、48 h、72 h和96 h半致死浓度分别为0.6040 mg/L、0.5148 mg/L、0.3812 mg/L和0.298 mg/L,安全质量浓度为0.02398 mg/L。肝脏和鳃中铜离子浓度随着水中铜离子浓度的增加而增加,且肝脏中的铜离子浓度小于鳃中;24 h时高浓度组(0.17 mg/L、0.30 mg/L、0.51 mg/L和0.90 mg/L)鱼肝脏中铜离子浓度与对照组差异显著(P<0.05),而鳃中高浓度组(0.30 mg/L、0.51 mg/L和0.90 mg/L)与对照组差异显著(P<0.05)。铜离子胁迫24 h时,花鳅肝中POD和GDH活性及脑中Glu浓度随水中铜离子浓度的增加而上升,不同组浓度组间显著差异(P<0.05);最高浓度组(0.9 mg/L)的活性最大,Glu浓度最高。本研究可为花鳅生态毒理学指标提供基础数据。The acute toxicity of copper ion to spined loach Cobitis taenia was conducted by way of static exposure.Juvenile spined loach with body weight of(4.1±0.6)g was exposed to copper sulfate(CuSO4)solution with Cu2+concentrations of 0 mg/L(control),0.10 mg/L,0.17 mg/L,0.30 mg/L,0.51 mg/L and 0.90 mg/L in a plastic 75 cm×60 cm×50 cm tank at stocking density of 30 fish per tank at water temperature of(15±1)℃.The contents of copper ion in gills and liver,and activities of peroxidase(POD),and glutamate dehydrogenase(GDH)in liver and glutamine content(Glu,μmol/g prot)in brain were determined in the spined loach exposed to the Cu2+concentrations above for 24 hour in order to explore the acute toxicity of copper ion to spined loach and to evaluate the safety of copper ion on spined loach.It was found that the toxic effect of Cu2+on spined loach was increased with increase in copper ion concentration,with median-lethal concentrations(LC50)of 0.6040 mg/L in 24 h,0.5148 mg/L in 48 h,0.3812 mg/L in 72 h and 0.298 mg/L in 96 h,and safe concentration of 0.02398 mg/L.The concentration of copper ion in the liver and gills were shown to be increased with the increase in copper ion concentration in the water,lower in the liver than that in the gills;significant difference in the liver between the high Cu2+concentration groups(0.17 mg/L,0.30 mg/L,0.51 mg/L and 0.90 mg/L)and the control at 24 h exposure(P<0.05),and significant difference in the gills between the high Cu2+concentration groups the high concentration groups(0.30 mg/L,0.51 mg/L,and 0.90 mg/L)and the control group at 24 h exposure(P<0.05).At 24 h of exposure,the activities of POD and GDH and Glu concentration were significantly increased with the increase in copper concentration in water,the maximal activity and Glu concentration in Cu2+concentration of 0.90 mg/L group.The finding provides the basic data for the ecotoxicology indices of spined loach C.taenia.
