摘  要:目的:观察电针对2型糖尿病大鼠血清炎性因子TNF-α、IL-6、IL-1β、空腹血糖(FBG)、胰岛素抵抗水平及骨骼肌IRS-1 Ser^307、GLUT4蛋白表达的影响,从而探讨电针足三里、三阴交、脾俞及胃脘下俞穴治疗2型糖尿病的可能机制。方法:以7只雄性ZL大鼠作为空白组,14只雄性ZDF大鼠随机分为2型糖尿病模型组和电针组(每组各7只)。干预4周,每周末检测空腹血糖,4周后取血清,放免法检测TNF-α、IL-6、IL-1β及胰岛素,取骨骼肌进行Western Blot检测IRS-1 Ser^307、GLUT4蛋白表达,并计算胰岛素抵抗相关指标。结果:电针组、模型组与空白组相比,TNF-α、IL-6、IL-1β水平均显著升高(P<0.05);电针组与模型组相比,TNF-α、IL-6、IL-1β水平均明显下降,差异有统计学意义(P<0.05);电针组、模型组与空白组相比,空腹血糖均显著升高(P<0.05);电针组与模型组相比,空腹血糖水平下降,但差异无统计学意义(P>0.05);电针组与空白组比较,骨骼肌IRS-1 Ser^307、GLUT4蛋白表达有所提高,但差异无统计学意义(P>0.05);电针组与模型组比较,骨骼肌IRS-1 Ser^307蛋白表达明显下降(P<0.05),而GLUT4蛋白表达明显提高(P<0.05);电针组、模型组与空白组相比,大鼠胰岛素抵抗指数水平明显提高(P<0.05);电针组与模型组相比,血清胰岛素抵抗指数水平差异有统计学意义(P<0.05)。结论:电针可调节2型糖尿病大鼠血清炎性因子、IRS-1 Ser^307、GLUT4蛋白表达,其可部分解释电针对IR以及2型糖尿病的作用机制及作用途径。Objective:To observe the effect of electro-acupuncture on serum inflammatory factors of TNF-α,IL-6,IL-1β,fasting blood glucose(FBG),insulin resistance and the protein expressions of IRS-1 Ser^307 and GLUT4 in skeletal muscle of type 2 diabetic rats,so as to explore the possible mechanism of electro-needling Zusanli(ST36),Sanyinjiao(SP6),Pishu(BL20)and Weiwanxiashu(EX-B3)in the treatment of type 2 diabetes.Methods:7 male ZL rats were used as the blank control,and 14 male ZDF rats were randomly divided into the model group and the electro-acupuncture group(with 7 rats in each group).After 4 weeks of intervention,fasting blood glucose was detected at the end of each week.Serum sample was collected and TNF-α,IL-6,IL-1βand insulin were detected by radioimmunoassay.The protein expressions of IRS-1 Ser^307 and GLUT4 in skeletal muscle were detected by Western Blot method,and insulin resistance related indexes were calculated.Results:Compared to the blank group,the levels of TNF-α,IL-6 and IL-1βwere significantly higher after modeling(P<0.05);after electro-acupuncture intervention,they were significantly lower compared to those in the model group(P<0.05).The fasting blood glucose and glucose were significantly higher in the electro-acupuncture group and the model group compared to those in the blank group(P<0.05);compared to the model group,the fasting blood glucose and glucose decreased in the electro-acupuncture group,but the difference was not statistically significant(P>0.05).Compared to the blank group,the protein expressions of IRS-1Ser^307 and GLUT4 in skeletal muscle were increased in the electro-acupuncture group,but there were no statistical difference between the electro-acupuncture group and the blank group(P>0.05).Compared to the model group,the expression of IRS-1 Ser^307 protein in skeletal muscle was significantly decreased(P<0.05),whereas the expression of GLUT4 protein was significantly increased in the electro-acupuncture group(P<0.05).Compared to the blank group,the level of insulin resistance i
