摘  要:目的:观察天麻素穴位注射对戊四氮(PTZ)致痫大鼠行为学及海马组织COX-2 mRNA表达的影响。方法:120只健康Wistar雄性大鼠随机分为空白组、模型组、穴注组及西药组,每组均为30只,采用戊四氮腹腔注射法进行癫痫模型造模,造模成功后治疗28 d,末次治疗结束时记录各组大鼠水迷宫、八臂迷宫试验结果,于迷宫试验结束后处死大鼠,取大鼠脑组织海马区,采用实时荧光定量PCR方法检测COX-2 mRNA表达量的变化。结果:除空白组大鼠外,其余各组大鼠均有痫性发作,且与模型组大鼠比较,穴注组及西药组癫痫大鼠的全面性强直-阵挛发作(GTCS)潜伏期和极小阵挛性发作(MCS)潜伏期均显著延长;Morris水迷宫试验示,模型组大鼠与空白组大鼠比较,定位航行游泳轨迹以随机式为主,定位航行所用时间明显延长,穿越平台数明显少于空白组;与模型组比较,穴注组及西药组大鼠定位航行游泳轨迹以直线式、趋向式为主,定位航行所用时间明显缩短,穿越平台数明显增多;八臂迷宫试验示,模型组大鼠与空白组大鼠比较,模型组大鼠参考记忆错误次数、工作记忆错误次数及吃完食物总时间多于空白组;与模型组比较,穴注组及西药组大鼠参考记忆错误次数、工作记忆错误次数及吃完食物总时间少于模型组;各组大鼠海马组织的COX-2 mRNA表达水平与空白组相比,模型组大鼠显著升高,穴注组、西药组大鼠表达升高;与模型组对比,穴注组、西药组干预后均降低COX-2 mRNA的表达水平。结论:天麻素穴位注射可明显延长癫痫大鼠GTCS潜伏期和MCS潜伏期,提示天麻素穴位注射有抗癫痫作用;穴注组大鼠在Morris水迷宫试验及八臂迷宫试验中学习记忆能力与模型组大鼠对比有明显增强,且穴注组COX-2 mRNA表达较模型组显著下降,提示天麻素穴注对戊四氮致痫大鼠的认知功能损害有改善作用,可能通过调节癫痫大鼠COX-2的�Objective:To observe the effect of acupoint injection with gastrodin on ethology and COX-2 mRNA expression of pentylenetetrazol-induced epileptic rats.Methods:120 healthy Wistar male rats were randomly divided into the blank group,the model group,the acupoint injection group and the western medication group,with 30 rats in each group.The epileptic model was established by intraperitoneal injection of pentylenetetrazol(PTZ).The treatment duration was 28 days.At the end of the last treatment,the results of water maze and eight-arm maze test were recorded.The hippocampus samples were collected after the maze tests.The change of COX-2 mRNA expression was detected by real-time fluorescence quantitative PCR.Results:Except for the blank group,the rats in all the other groups had seizures,and compared to the model group,acupoint injection group and western medication group could significantly prolong the latencies of generalized tonic-clonic seizure(GTCS)and minimal clonic seizure(MCS)in epileptic rats.In terms of the Morris water maze test,it showed that the positioning swimming trajectory of the model group was mainly random and the navigation time was significantly prolonged compared to those of the blank group,and the number of crossing platforms was significantly less than that of the blank group;compared to the model group,the positioning swimming trajectory was mainly linear and trending in the acupoint injection group and the western medication group,the navigation time was obviously shortened,and the number of crossing platforms was significantly increased.In terms of the eight-arm maze test,it showed that compared to the blank group,the number of reference memory errors and working memory errors were more and the total time of eating food was longer in the model group than those in the blank group;the number of reference memory errors and working memory errors were less and the total time of eating food was shorter in the acupoint injection group and western medication group than those in the model group.Compar
