摘  要:目的:研究电针对骨骼肌萎缩模型大鼠的干预效果及作用机制。方法:30只大鼠随机分为模型组、假手术组和电针组,各10只,建立骨骼肌萎缩模型。电针组给予电针治疗,假手术组和模型组大鼠每天进行固定。检查腓肠肌湿重、干重,骨骼肌肌纤维直径、面积,采用ELISA检测泛素蛋白连接酶肌肉环指蛋白1(MURF1)、PGC-1α、Ⅲ型纤连蛋白组件包含蛋白5(FNDC5)蛋白,采用Western Blot检测Akt、mTOR、p-Akt及p-mTOR。结果:电针组骨前肌湿重、干重、干重/体重、骨骼肌肌纤维直径和面积高于模型组,低于假手术组(P<0.05);模型组骨前肌湿重、干重、干重/体重、骨骼肌肌纤维直径和面积低于假手术组(P<0.05)。电针组骨骼肌相关蛋白MURF1表达低于模型组,PGC-1α、FNDC5表达高于模型组;电针组骨骼肌相关蛋白MURF1表达高于假手术组,PGC-1α、FNDC5表达低于假手术组(P<0.05);模型组骨骼肌相关蛋白MURF1表达高于假手术组,PGC-1α、FNDC5表达低于假手术组(P<0.05)。电针组Akt、mTOR、p-Akt及p-mTOR表达高于假手术组和模型组(P<0.05);模型组Akt、mTOR、p-Akt、p-mTOR表达低于假手术组(P<0.05)。结论:电针对骨骼肌萎缩模型大鼠干预效果显著,能改善大鼠骨骼肌萎缩状况,通过激活Akt/mTOR通路,调控骨骼肌相关蛋白MURF1、PGC-1α和FNDC5表达,从而抑制骨骼肌萎缩。Objective:To study the intervention effect and mechanism of electro-acupuncture on skeletal muscle atrophy model rats.Methods:30 rats were randomly divided into the model group,the sham-operation group and the EA group,with 10 rats in each group.Skeletal muscle atrophy model was established.The rats in the EA group were treated with electro-acupuncture,and the rats in the sham-operation group and the model group were fixed at the same time point everyday.Wet and dry weight of gastrocnemius muscle,diameter and area of skeletal muscle fibers were examined.The expressions of MURF1,PGC-1αand FNDC5 were detected by ELISA method.The expressions of Akt,mTOR,p-Akt and p-mTOR were detected by Western Blot method.Results:The wet weight,dry weight,dry weight/body weight,diameter and area of skeletal muscle fibers were higher in the EA group than those in the model group but lower than those in the sham-operation group(P<0.05).The wet weight,dry weight,dry weight/body weight,diameter and area of skeletal muscle fibers were lower in the model group than those in the sham-operation group(P<0.05).The expression of MURF1 was lower and the expressions of PGC-1αand FNDC5 were higher in the EA group than those in the model group(P<0.05).The expression of MURF1 was higher,and the expressions of PGC-1a and FNDC5 were lower in the EA group than those in the sham-operation group(P<0.05).The expression of MuRF1 was higher and the expressions of PGC-1αand FNDC5 were lower in the model group than those in the sham-operation group(P<0.05).The expressions of Akt,mTOR,p-Akt and p-mTOR were higher in the EA group than those in the sham-operation group and the model group(P<0.05).The expressions of Akt,mTOR,p-Akt and p-mTOR were lower in the model group than those in the sham operation group(P<0.05).Conclusion:The intervention effect of electro-acupuncture on skeletal muscle atrophy model rats is remarkable.It can improve the condition of skeletal muscle atrophy in rats.By activating the Akt/mTOR pathway,it can regulate the expressions of M
