复方乌鳢口服液对产后抑郁症模型大鼠行为学的影响

摘　　要：目的:观察复方乌鳢口服液对产后抑郁症(PPD)模型大鼠行为学的影响及其作用机制。方法:60只SPF级受孕SD大鼠(体质量250~280g)随机分为对照组、模型组、阳性对照组、复方乌鳢口服液低、中、高剂量组。观察母鼠对幼崽的关爱行为,通过旷场实验、悬尾实验、强迫游泳实验,研究复方乌鳢口服液抗抑郁作用,并测定母鼠血清中5-羟色胺(5-HT)含量。结果:与空白对照组比较,模型组大鼠对幼崽关爱行为减少,悬尾实验及强迫游泳实验大鼠绝望不动时间延长,旷场实验大鼠活动性减少,血清5-HT含量降低(P<0.05);与模型组大鼠比较,复方乌鳢口服液组和阳性对照组大鼠对幼崽关爱行为增多,悬尾实验及强迫游泳实验大鼠绝望不动持续时间减少,旷场实验大鼠活动性及兴趣性增加,血清5-HT含量升高(P<0.05)。结论:复方乌鳢口服液能够改善PPD模型大鼠的行为学指标及升高血清5-HT含量,具有抗抑郁作用。Objective:To observe the effect of Compound Snakehead Oral Liquid on the behavior of rats with postpartum depression(PPD)and its mechanism.Method:60 SPF conceived SD rats(body weight 250-280 g)were randomly divided into control group,model group,positive control group,and low,medium and high dose groups of Compound Snakehead Oral Liquid.The caring behavior of mother rats to their pups,through open field experiment,tail suspension experiment,forced swimming experiment,the antidepressant effect of Compound Snakehead Oral Liquid was studied,and the serum serotonin(5-HT)content of mother rats was determined.Result:Compared with the blank control group,rats in the model group had less caring behaviors for their pups.The rats in the model group had a longer desperate immobility time in the tail suspension experiment and the forced swimming experiment.The rats in the model group had less activity in the open field experiment.The serum 5-HT content in the model group decreased(P<0.05).Compared with the model group rats,the rats in the Compound Snakehead Oral Liquid group and the positive control group have more caring behaviors for the pups.The rats in the Compound Snakehead Oral Liquid group and the positive control group have less desperate immobilityin the tail suspension experiment and the forced swimming experiment.The activity and interest of rats in the Compound Snakehead Oral Liquid group and the positive control group increased in the open field experiment.The serum 5-HT content increased in the Compound Snakehead Oral Liquid group and the positive control group(P<0.05).Conclusion:Compound Snakehead Oral Liquid could improve the behavioral indicators of PPD model rats and increase the serum 5-HT content,with antidepressant effect.
