高效液相色谱法测定大菟丝子中咖啡酸的含量

摘　　要：目的:采用高效液相色谱法测定民族药材大菟丝子中的咖啡酸含量。方法:以乙腈-0.05%磷酸溶液(10∶90)为流动相,流速1.0 mL/min,检测波长327 nm,采用C18色谱柱,柱温30℃,外标法定量。结果:咖啡酸在0.265~10.6μg/mL浓度范围内与峰面积呈良好的线性关系(r=0.9992),平均回收率为99.9%(RSD=1.8%)。结论:该测定方法快速、简便、专属性较强,可用于大菟丝子中咖啡酸含量的测定和质量控制。Objective:Establish a method for determination of caffeic acid in Cuscuta japonica Choisy with HPLC.Methods:The mobile phase was consisted of acetonitrile-0.05%phosphoric acid aqueous solution(10∶90),The flow rate was 1.0 mL/min,the detective wavelength was 327 nm and the C18 column temperature was 30℃.With external standard method to make quantitative analysis.Results:The linear range of the method was 0.265~10.6μg/mL,the average recovery was 99.9%,RSD was 1.8%.Conclusion:This method is simple,fast and efficient,which will be used to provide accurate,scientific quality control for determination of caffeic acid in Cuscuta japonica Choisy.
