摘  要:探讨藏药垂头菊体外抗肿瘤作用,为民族药开发利用提供科学依据。本研究以垂头菊黄酮为研究材料,小鼠S180、Hepa1-6、L1210、B16-F10、SP2/0瘤细胞株为研究对象,CCK-8、AO/EB、Annexin V/PI流式细胞术为研究手段,细胞生长抑制率、细胞凋亡形态和凋亡率为检测指标,研究藏药垂头菊黄酮体外抗肿瘤作用。研究结果表明:不同浓度垂头菊黄酮对L1210、S180、Hepa1-6、B16-F10瘤细胞的生长呈现不同的抑制作用;L1210瘤细胞为垂头菊黄酮敏感细胞,诱导L1210瘤细胞凋亡作用具有剂量依赖性特点,浓度在500~750μg/mL之间具有与阳性药物5-FU相当的诱导瘤细胞凋亡作用,浓度在1000μg/mL时,具有最大的细胞凋亡率或死细胞率(98.74%)。To explore the antitumor effect of Tibetan medicine cremanthodium chinensis in vitro and provide scientific basis for the development and utilization of ethnic medicine.In this study,the flavonoids of cremanthodium discoideum maxim were used as research materials,and the mouse S180,Hepa1-6,L1210,B16-F10,SP2/0 tumor cell lines were used as research objects,CCK-8,AO/EB,Annexin V/PI flow cytometry as research methods,cell growth inhibition rate,apoptotic morphology,and apoptotic rate are indicators of detection,study on antitumor effect of Tibetan medicine cremanthodium flavone in vitro.The results showed that:different concentrations of flavonoids of cremanthodium showed different inhibitory effects on the growth of L1210,S180,Hepa1-6,and B16-F10 tumor cells;L1210 tumor cells are cremanthodium flavonoid sensitive cells,which induce the apoptosis of L1210 tumor cells in a dose-dependent manner.The concentration of L1210 tumor cells is equivalent to that of the positive drug 5-FU at concentrations between 500 and 750μg/mL.At 1000μg/mL,it has the largest rate of apoptosis or dead cells(98.74%).
