摘 要:旨在研究镉对小鼠腹腔巨噬细胞的毒性损伤及锌对镉染毒巨噬细胞免疫损伤的拮抗作用。提取雄性ICR小鼠腹腔巨噬细胞,建立体外镉染毒(20、50、100μmol/L)和锌补充(20μmol/L)模型,分别对巨噬细胞形态学、吞噬功能和炎性细胞因子表达进行检测。另外,通过饮水方式对小鼠进行镉攻毒(25、250μmol/L)和锌补充(220μmol/L),3个月后提取各组小鼠腹腔巨噬细胞,观察其形态变化和吞噬活性。结果显示,镉染毒造成小鼠腹腔巨噬细胞浓度依赖性形态和超微结构损伤,细胞吞噬功能下降和LPS诱导的炎性细胞因子(IL-1β、TNF-α和IL-6)表达量降低。锌补充能显著提高各浓度镉染毒小鼠腹腔巨噬细胞的吞噬能力,不同程度增强轻度(20μmol/L)和中度(50μmol/L)镉染毒巨噬细胞LPS诱导的IL-1β、TNF-α和IL-6基因表达(P<0.05),但对高浓度(100μmol/L)镉染毒巨噬细胞免疫保护作用不明显。结果表明,镉染毒造成小鼠腹腔巨噬细胞浓度依赖性细胞毒性和免疫功能损伤,锌补充能显著增强镉染毒小鼠巨噬细胞的吞噬活性,恢复轻度、中度镉染毒巨噬细胞炎性细胞因子的表达水平。The present study was to investigate the toxic damage of cadmium to mouse peritoneal macrophages and the inhibitory effect of zinc on the cadmium-caused immune function damage to mouse peritoneal macrophages.The peritoneal macrophages of male ICR mice were extracted,cadmium poisoning(20,50,100μmol/L)and/or zinc supplement(20μmol/L)models were established in vitro.After the cadmium and/or zinc treatment,morphology,phagocytic function and expression of inflammatory cytokines in mouse peritoneal macrophages were detected respectively.In addition,cadmium poisoning(25,250μmol/L)and/or zinc supplement(220μmol/L)models were established in vivo by oral administration.After treatment for three months,morphological changes and phagocytic activity of the peritoneal macrophages in each group were detected.In result,cadmium exposure caused dose-dependent morphological and ultrastructural damage to mouse peritoneal macrophages,decreasing the cell phagocytosis and expression of LPSinduced inflammatory cytokines(IL-1β,TNF-αand IL-6).Zinc supplement significantly increased the phagocytic capacity of peritoneal macrophages in cadmium-exposed mice of various concentrations,and enhanced the LPS-stimulated expressions of IL-1β,TNF-αand IL-6 in mild and moderate cadmium-exposed(20,50μmol/L)mouse peritoneal macrophages(P<0.05).However,the immunoprotection of zinc on peritoneal macrophages exposed to high dose of cadmium(100μmol/L)was not obvious.In conclusion,cadmium exposure led to dose-dependent cytotoxicity and immune damage to mouse peritoneal macrophages.Zinc supplement significantly enhanced the phagocytosis of cadmium-exposed mouse macrophages and can restore the expression of inflammatory cytokines in mildly and moderately cadmium-exposed macrophages.
