摘 要:为了探寻连续移植对强致病力层出镰孢菌株遗传变异的影响,本试验利用生物学测定和ISSR分子标记对层出镰孢菌连续移植20次菌株的菌落形态、显微形态、致病性和遗传多样性等进行了研究。生物学测定结果表明:不同移植次数菌株在菌落形态和菌体形态上未表现出明显差异;但菌丝生长速率、产孢量和致病力随着移植次数增多而表现出下降趋势,并且菌株产生菌核的能力减弱。ISSR分子标记分析结果表明:层出镰孢菌连续移植菌株间基因组DNA在简单重复序列间区存在丰富的遗传多样性;虽然大多数移植次数相近的菌株间表现出较高的遗传相似性,但菌株间的遗传分化明显,而这种遗传分化与菌株间表现出的生物学特性无明显的相关性。综上所述,层出镰孢菌株经过连续移植后出现了显著的退化现象。To explore the influence of serial transplantation on genetic variation of Fusarium proliferatum with high pathogenicity,we studied the colonial morphology,microscopic morphology,pathogenicity and genetic diversity of strains in 20 serially transplanted times by the biology assays and ISSR molecular markers.The biology assay results showed that all the serially transplanted strains did not appear obvious differences in colony morphology and micro-morphology,while the mycelia growth rate,sporulation rate and pathogenicity appeared declining trend with the increase of transplanted times.The generation capability of sclerotium was weakened.The ISSR molecular markers analysis results indicated that there were abundant genetic diversity of the ISSR region among different serially transplanted F.proliferatum.Although most of the adjacently transplanted strains appeared relatively high genetic similarity,there was obvious genetic differentiation.There was no obvious correlation between the tested biological characteristics and genetic differentiation.To sum up,there was obvious degeneration of the serially transplanted F.proliferatum.
